摘 要:目的研究不同浓度MT浸种对Cu胁迫下远志种子萌发、幼苗生长及类黄酮代谢的影响,探明MT缓解Cu胁迫的最适浓度。方法采用培养皿发芽法,研究了不同浓度MT浸种(0、5、25、50、100、200、400μmol/L)对Cu胁迫下远志种子萌发、幼苗生长、MDA含量、类黄酮含量及其合成关键酶活性的影响。结果不同浓度Cu胁迫对远志种子萌发和幼苗生长产生“低促高抑”的影响。与100 mg/L Cu胁迫相比,100μmol/L MT浸种能有效提升Cu胁迫下远志种子的发芽势、发芽率、发芽指数、活力指数以及幼苗的苗高、根长、鲜重、干重、类黄酮含量和PAL、4CL、C4H活性(P<0.05),而明显降低MDA含量(P<0.05)。结论适宜浓度的MT浸种能明显提升远志的抗氧化能力,从而减轻Cu胁迫对种子萌发和幼苗生长产生的抑制。Objective To study the effects of seed soaking with different concentrations of MT on seed germination,seedling growth and flavonoids metabolism of Polygala tenuifolia under Cu stress,and to explore the optimal MT concentration to alleviate Cu stress.Methods A culture dish germination method is conducted to study the effects of seed soaking with different concentrations of MT(0,5,25,50,100,200,400μmol/L)on seed germination,seedling growth,MDA content,flavonoids content and flavonoids synthetic key enzyme activities of Polygala tenuifolia under Cu stress.Results Seed germination and seedling growth under Cu stress all show effects of“low-promotion,high-inhibition”.Compared with 100 mg/L Cu stress,MT seed soaking at 100μmol/L can effectively improve the germination potential,germination rate,germination index and vigor index,seedling height,root length,fresh weight,dry weight,flavonoid content and the PAL,4 CL and C4 H activities of Polygala tenuifolia under Cu stress(P<0.05),but significantly reduce the MDA content(P<0.05).Conclusion MT seed soaking at an appropriate concentration can significantly improve the antioxidant capacity of Polygala tenuifolia,so as to reduce the inhibition of Cu stress on seed germination and seedling growth.
