摘  要:目的通过对广州人工湖水域夏冬两季水样进行18S rDNA测序分析,寻找不同人工湖的硅藻及其他真核浮游生物分布和差异,从而为溺死诊断和落水地点的推断提供依据。方法在3个人工湖(白云湖、东山湖和海珠湖)中各选取等距离的3个取水点,分别在夏冬两季采集水样,采用18S rDNA测序对各取水点的硅藻及其他真核浮游生物进行定性和定量分析。结果夏冬两季各人工湖中包括硅藻在内的真核浮游生物的种类数量具有显著差异,分别为477.22±25.12个和284.48±13.79个。而不同人工湖之间的硅藻含量具有显著的时空分布差异。此外,各人工湖硅藻的种类和数量不完全相同,其中白云湖特有硅藻种属为Fistulifera属、拟菱形藻属、肘杆藻属和Roundia属,而东山湖特有种属包括双眉藻属、异极藻属和布纹藻属,海珠湖仅为脆杆藻属。结论对于水中尸体的法医学分析,不同人工湖的真核浮游生物群落分布及其差异可为下一步建立水中真核浮游生物种群的背景数据库积累基础数据,进而为死因和溺死地点的推断提供参考。Objective To explore the distribution of diatoms and other eukaryotic plankton in different artificial lakes and to provide a basis for the diagnosis of drowning and the inference of the location of falling water,we performed 18S rDNA sequencing on the water samples of the artificial lakes in Guangzhou in summer and winter.Methods Water samples were collected at 3 equidistant water intake points in each of the three artificial lakes(Baiyun Lake,Dongshan Lake and Haizhu Lake)in summer and winter.Subsequently,18S rDNA sequencing was used to qualitatively and quantitatively analyze the diatoms and other eukaryotic plankton in each sample.Results There were significant differences in the number of eukaryotic plankton species(including diatoms)in the artificial lakes between summer and winter,which were 477.22±25.12 and 284.48±13.79,respectively.The diatom abundance among the different artificial lakes had significant temporal and spatial distribution differences.In addition,the types and numbers of the diatoms in the artificial lakes were not consistent.Among them,the endemic diatom species were Fistulifera,Pseudo-nitzschia,Ulnaria and Roundia in the Baiyun Lake,while were Amphora,Gomphonema and Gyrosigma in the Dongshan Lake.The Haizhu Lake had only Fragilaria.Conclusion For the forensic analysis of the corpses in water,the distribution data of eukaryotic plankton communities in different artificial lakes can be used for the establishment of a background database of eukaryotic plankton populations in water,and then provide evidences for inferring the cause of death and the location of drowning.
