摘 要:附子的强心作用机制是近年的研究热点之一。药理学研究发现附子可通过减少心肌细胞凋亡、调节异常的神经内分泌系统等发挥强心作用,其作用机制包括调节心肌细胞内Ca^(2+)超载,调节超氧化物歧化酶、过氧化氢酶等相关蛋白及AMPK/mTOR、PI3K/Akt信号通路,选择性激动肾上腺素受体抑制心肌细胞凋亡等病理过程,调节BNP、TNF-α、ANP等神经内分泌因子与炎症细胞因子水平异常,促进心室重构等。本研究通过检索国内外相关文献,对附子发挥强心作用的分子机制进行了系统综述,以期为附子的临床应用和强心药物研发提供理论依据。In recent years,the cardiotonic mechanism of Fuzi(Aconiti Lateralis Radix Praeparata)has always been one of the research hotspots.Modern pharmacological studies have found that Fuzi can exert cardiotonic effect by reducing myocardial cell apoptosis,regulating abnormal neuroendocrine system,and etc.Its mechanism of action includes regulating Ca^(2+) overload in cardiomyocytes,regulating superoxide dismutase,catalase,and other related proteins,and AMPK/mTOR,PI3K/Akt signaling pathway,selectively stimulate adrenergic receptors to inhibit cardiomyocyte apoptosis and other pathological processes,regulate the levels of abnormal neuroendocrine factors and inflammatory cytokines such as BNP,TNF-α,ANP,and promote ventricular remodeling.This study systematically reviewed the molecular mechanism of Fuzi’s cardiotonic effect by searching relevant literatures at home and abroad,in order to provide theoretical basis for clinical application of Fuzi and research and development of cardiotonic drugs.
