摘 要:为了稳定半枫荷增殖、增加有效的组培苗、提高生根率,采用改变继代培养方式、培养条件等措施,进行半枫荷组培生产试验。结果表明,选择春季萌发的枝条为外植体,培养基为MS+6-BA 1.0 mg/L+NAA 0.2 mg/L+蔗糖30 g/L有利于诱导芽;培养基为改良MS+6-BA 1.2 mg/L和NAA 0.3 mg/L,并在5000±200 Lx的光照条件下培养,可明显改善玻璃化现象,有利于芽的增殖;培养基为1/2MS+IBA 0.8 mg/L+NAA 0.4 mg/L+蔗糖20 g/L+活性炭0.2 g/L有利于根的诱导;直接用泥炭土作为基质,有利于组培苗的移栽。In order to stabilize propagation of Semiliquidambar cathayensis,increase the effective tissue culture plantlets and improve the rooting rate,such measures as changing the subculture mode and culture conditions were adopted in producing the tissue culture plantlets.The results showed that using the spring shoots as explants and the culture medium MS+6-BA 1.0 mg/L+NAA 0.2 mg/L+sucrose 30 g/L could facilitate bud induction;the culture condition with the improved MS+6-BA 1.2 mg/Land NAA 0.3 mg/L as the culture medium and lighting of 5000±200 Lx could significantly eliminate vitrification and promote propagation;the culture medium 1/2MS+IBA 0.8 mg/L+NAA 0.4 mg/L+sucrose 20 g/L+activated charcoal 0.2 g/L could facilitate root induction;the culture medium of pure peat soil could facilitate transplantation of the tissue culture plantlets.
