摘 要:目的探讨九龙藤总黄酮(BCF)通过lncRNA心肌梗死相关转录本(MIAT)对大鼠急性心肌梗死(AMI)的作用及机制。方法随机选取雄性SD大鼠,分为sham组、AMI组、BCF组,除假手术组外,其余各组采用左冠状动脉前降支结扎法制备大鼠心肌缺血模型,缺血24 h后进行lnc RNA基因检测;设计并合成lnc RNA MIAT si-RNA,将雄性SD大鼠分为si-NC组、siMIAT组、BCF+si-NC组和BCF+si-MIAT组,按实验分组制备大鼠心肌缺血模型,随后HE染色观察心脏组织病理学变化,ELISA实验检测血清心肌钙蛋白Ⅰ(cTn-Ⅰ)含量,透射电镜观察自噬小体数量,RT-qPCR和Western blot检测lncRNA MIAT、Beclin1、Cathepsin D与LC3表达。结果BCF+AMI组MIAT表达低于AMI组(P<0.05)。HE染色结果显示,BCF+siMIAT组心肌细胞形态较BCF+si-NC组进一步改善;AMI+si-MIAT组、AMI+BCF+si-NC组和AMI+BCF+si-MIAT血清cTn-Ⅰ含量低于AMI+si-NC组(P<0.05),且AMI+BCF+si-MIAT组血清cTn-Ⅰ含量低于AMI+BCF+si-NC组(P<0.05);BCF+si-MIAT组自噬小体数量低于BCF+si-NC组(P<0.05);AMI+si-NC组Beclin1、Cathepsin D、LC3表达及蛋白高于AMI+si-MIAT组、AMI+BCF+si-NC组、AMI+BCF+si-MIAT组(P<0.05);而AMI+BCF+si-MIAT组Beclin1、Cathepsin D、LC3表达低于AMI+BCF+si-NC组(P<0.05)。结论BCF可能通过MIAT抑制自噬抗AMI。Objective To investigate the effect and mechanism of total flavonoids of Bauhinia Championii flavone(BCF)on acute myocardial infarction(AMI)in rats through lncRNA myocardial infarction related transcript(MIAT).Methods Male SD rats were randomly selected and divided into sham group,AMI group,si-NC group,si-MIAT group,BCF group,except for sham group,myocardial ischemia model was prepared by ligation of anterior descending branch of left coronary artery in other groups.lnc RNA gene was detected 24 hours after ischemia.lnc RNA MIAT si-RNA was designed and synthesized.Male SD rats were divided into si-NC group,si-MIAT group,BCF+si-NC group and BCF+si-MIAT group.Rat myocardial ischemia model was prepared by experimental groups.Then the hipathological changes of heart were observed by HE staining,the level of serum cTn-Ⅰ was detected by ELISA assay,the number of autophagosomes was viewed with transmission electron microscopy,and the expression levels of MIAT,Beclin1,Cathepsin D and LC3 were detected by RT-qPCR and Western blot.Results The expression of MIAT in BCF+AMI group was lower than that in AMI group(P<0.05).HE staining results showed that the morphology of cardiomyocytes in the BCF+si-MIAT group was further improved than that in the BCF+si-NC group.The serum cTn-I content of AMI+si-MIAT group,AMI+BCF+si-NC group and AMI+BCF+si-MIAT group was lower than that of AMI+si-NC group(P<0.05),and the serum cTn-I content of AMI+BCF+si-MIAT group was lower than that of AMI+BCF+si-NC group(P<0.05).The number of autophagosomes in BCF+si-MIAT group was lower than that in BCF+si-NC group(P<0.05).The expression and protein of Beclin1,Cathepsin D and LC3 in AMI+si-NC group were higher than those in AMI+si-MIAT group,AMI+BCF+si-NC group and AMI+BCF+siMIAT group(P<0.05),and the expression of Beclin1,Cathepsin D and LC3 in AMI+BCF+si-MIAT group was lower than that in AMI+BCF+si-NC group(P<0.05).Conclusion BCF may inhibit autophagy and anti-AMI by MIAT.
