摘 要:目的:建立延胡索与醋延胡索HPLC指纹图谱,并结合化学计量学分析研究,为全面评价延胡索的质量提供参考。方法:采用HPLC法测定。色谱柱为Agilent XDB C_(18)(250 mm×4.6 mm,5μm),流动相为乙腈-0.1%磷酸水溶液,梯度洗脱,检测波长280 nm。采用“中药色谱指纹图谱相似度评价系统”(2012A版)建立延胡索和醋延胡索HPLC指纹图谱;并对其中6个化合物含量测定方法进行方法学验证。采用偏最小二乘-判别分析(PLS-DA)化学计量学方法对延胡索和醋延胡索指纹图谱进行分析。结果:延胡索指纹图谱标定了17个共有峰,相似度为0.934~0.985,醋延胡索指纹图谱标定20个共有峰,相似度为0.954~0.987,指认其中6个共有峰分别为原阿片碱、盐酸巴马汀、盐酸小檗碱、脱氢紫堇碱、延胡索乙素、延胡索甲素,6个成分在各自的质量浓度范围内线性关系良好(r^(2)>0.999),平均加样回收率为100.2%~104.7%,RSD为0.12%~2.2%;PLS-DA将30批样品聚为2类,并分析出10个差异标志物。结论:建立了延胡索和醋延胡索指纹图谱及6个化合物含量测定方法,可为延胡索的炮制及质量控制提供参考。Objective:To establish the HPLC fingerprint of Corydalis Rhizoma and vinegar Corydalis Rhizoma,and to provide reference for comprehensive evaluation of the quality by chemometric method.Methods:The samples were determined by HPLC.An Agilent XDB C_(18) column(250 mm×4.6 mm,5 μm) was used with acetonitrile-0.1% phosphoric acid solution(gradient elution) as the mobile phase,and the detection wavelength was 280 nm.The HPLC fingerprints of Corydalis Rhizoma and vinegar Corydalis Rhizoma were established by using “The similarity evaluation system of traditional Chinese medicine chromatographic fingerprint”(2012 A version),and determination method of six compounds was validated.The fingerprints were analyzed by PLS-DA.Results:There were 17 and 20 common peaks in the fingerprints of Corydalis Rhizoma and vinegar Corydalis Rhizoma,respectively.The similarities were 0.934-0.985 and 0.954-0.987,respectively.Six compounds were identified as protopine,palmatine hydrochloride,berberinehydrochloride,dehydrocorydaline,tetrahydropalmatine and corydaline by comparison with the reference substances.The linear relationship of the six components in the range of their respective mass concentration was good(r~2>0.999),the average recovery of addition were 100.2%-104.3% and the RSDs were 0.94%-1.8%.30 samples were classified into two groups and 10 components were selected as the main differential markers by PLS-DA.Conclusion:The fingerprints of Corydalis Rhizoma and vinegar Corydalis Rhizoma and the content determination method of six compounds are successfully established,which can provide reference for processing and quality control of Corydalis Rhizoma.
