摘  要:目的探讨宝藿苷Ⅰ对1-甲基-4-苯基吡啶离子(MPP^(+))诱导SH-SY5Y人神经母细胞瘤细胞损伤保护作用及雌激素受体(ER)特异性阻断剂ICI 182,780的阻断效应。方法将SH-SY5Y细胞接种于96孔板,用不同浓度(0.01、0.10、1.00、10.00、20.00μmol/L)宝藿苷Ⅰ预保护24 h,加1 mmol/L MPP^(+)作用24 h,用四甲基偶氮唑蓝(MTT)法检测细胞存活率。将SH-SY5Y细胞分为对照组、MPP^(+)组、MPP^(+)+宝藿苷Ⅰ组、MPP^(+)+宝藿苷Ⅰ+ICI 182,780组和宝藿苷Ⅰ组,用免疫印迹法检测各组细胞α-突触核蛋白表达。结果0.10、1.00μmol/L宝藿苷Ⅰ能够对抗MPP^(+)诱导的SH-SY5Y细胞存活率降低,而10.00和20.00μmol/L宝藿苷Ⅰ会加重细胞的损伤(F=51.070,q=5.453~13.650,P<0.01)。与对照组比较,MPP^(+)组α-突触核蛋白表达明显上调(F=7.913,q=5.709,P<0.01);0.10μmol/L宝藿苷Ⅰ预保护能明显降低MPP^(+)诱导的α-突触核蛋白表达上调(q=3.485,P<0.05),此作用可以被ICI 182,780阻断(q=4.350,P<0.05);单独应用宝藿苷Ⅰ对α-突触核蛋白的表达无明显影响。结论宝藿苷Ⅰ能够抑制MPP^(+)诱导的SH-SY5Y细胞损伤,其机制可能与ER信号途径有关。Objective To investigate the protective effect of baohuosideⅠagainst human neuroblastoma SH-SY5Y cell damage induced by 1-methyl-4-phenylpyridinium(MPP^(+))and the blocking effect of ICI 182,780,a specific estrogen receptor(ER)blocker.Methods SH-SY5Y cells were inoculated into 96-well plates,pre-protected with different concentrations of baohuosideⅠ(0.01,0.10,1.00,10.00,and 20.00μmol/L)for 24 h,and then treated with 1 mmol/L MPP^(+) for another 24 h,and MTT assay was used to measure cell viability.SH-SY5Y cells were divided into control group,MPP^(+) group,MPP^(+)+baohuosideⅠgroup,MPP^(+)+baohuosideⅠ+ICI 182,780 group,and baohuosideⅠgroup,and Western blotting was used to measure the protein expression ofα-synuclein in each group.Results BaohuosideⅠat the concentrations of 0.10 and 1.00μmol/L exerted a protective effect against the MPP^(+)-induced reduction in SH-SY5Y cell viability,while baohuosideⅠat the concentrations of 10.00 and 20.00μmol/L aggravated cell damage(F=51.070,q=5.453-13.650,P<0.01).Compared with the control group,the MPP^(+) group had a significant increase in the protein expression ofα-synuclein(F=7.913,q=5.709,P<0.01),while pre-protection with 0.10μmol/L baohuosideⅠsignificantly inhibited the upregulation of α-synuclein induced by MPP^(+)(q=3.485,P<0.05),which was blocked by ICI 182,780(q=4.350,P<0.05),and baohuosideⅠtreatment alone had no significant effect on the protein expression ofα-synuclein.Conclusion BaohuosideⅠcan inhibit MPP^(+)-induced SH-SY5Y cell damage,which may be associated with the ER signaling pathway.
