摘 要:[目的]单宁是谷类粮食作物高粱的重要次生代谢物,其含量的高低直接影响高粱的用途。高粱调控单宁含量基因Tan1,是一个含有WD-40结构域的转录因子,通过调控单宁合成途径的关键酶而影响高粱单宁含量。Tan1基因的不同变异直接影响单宁含量,开发实用的分子标记对鉴定Tan1不同等位基因型颇为重要。[方法]本研究首先对4种已知等位基因型Tan1、tan1-a、tan1-b和tan1-c编码区序列和籽粒单宁含量进行关联分析;其次通过检测290份栽培种中Tan1基因的等位变异和单宁含量,评价4种等位基因型在育种中的应用情况;最后通过分析不同等位基因型的编码区序列开发Tan1基因分子标记便于快速鉴定。[结果]Tan1基因的3种变异类型由于编码区序列的插入、缺失、突变导致Tan1蛋白功能丧失造成高粱籽粒无单宁或者低单宁;无单宁或低单宁栽培种中tan1-a和tan1-b应用广泛,而tan1-c应用较少;基于扩增片段大小以及酶切开发了Tan1基因4种等位基因型的鉴定方法。[结论]本研究基于Tan1不同等位变异开发实用的分子标记,为无单宁或低单宁杂交种培育提供便捷的分子检测方法。[Objective]Tannin is an important secondary metabolite of grain crop sorghum,and its content directly affects the use of sorghum.Tan1 is a transcription gene containing a WD-40 domain and responsible for controlling the tannin content by regulating key enzymes in the tannin synthesis pathway in sorghum.Different variants of Tan1 gene directly affect tannin content,so it is important to develop practical molecular markers to identify different Tan1 alleles.[Methods]In this study,the correlation analysis of the coding region sequences and grain tannin contents of four known alleles,Tan1,tan1-a,tan1-b and tan1-c were analyzed first.Then,allelic variation and tannin contents in 290 cultivars were detected to evaluate the application of four Tan1 alleles in breeding.Finally,molecular markers of Tan1 were developed by analyzing the coding region sequences of different alleles for rapid identification.[Results]The three variants of Tan1 gene showed non-tannin or low-tannin in sorghum grains due to the insertion,deletion and mutation of the coding sequence.In non-tannin or low-tannin cultivars,tan1-a and tan1-b were widely used,while tan1-c was less used.Based on the size of amplified fragments and restriction enzyme digestion,a method for identifying four Tan1 alleles were developed.[Conclusion]Practical molecular markers were developed based on the different allelic variation of Tan1 which provide a convenient molecular detection method for the breeding of non-tannin or low-tannin hybrids.
