摘  要:为筛选维生素E及组分含量特异种质资源,研究以327份大豆种质资源为材料,采用改良的高效液相色谱法(HPLC),对大豆籽粒中的维生素E及其组分α-生育酚、γ-生育酚、δ-生育酚的含量进行测定。结果表明,利用改良的高效液相色谱法(HPLC)测定大豆维生素E含量精密度高,可靠性好;大豆种质资源中,维生素E及各生育酚含量的变异系数均大于20%。α-生育酚、总维生素E的含量呈现正偏态分布,γ-生育酚、δ-生育酚的含量呈正态分布;α-生育酚、γ-生育酚、δ-生育酚和总维生素E的含量之间都存在极显著相关关系。对种质资源各组分进行聚类分析,结果筛选出富含维生素E的大豆品种34个。热图分析表明,富含维生素E的不同种质各生育酚组分含量存在差异,筛选出α-生育酚含量较高的2份种质资源东北黑豆和山东平邑豆,γ-生育酚含量较高的3份种质资源SCS-9299、辽豆22和朔州平鲁双色豆,3种生育酚含量都较高的5份种质资源富黑2号、黑珍珠、于秀芝翠绿、特大粒黑皮绿豆、青仁黑一号。In order to screen special germplasm resources of vitamin E component, in this study, 327 soybean germplasm resources were taken as materials, the content of vitamin E and its components α-tocopherol, γ-tocopherol, and δ-tocopherol in the seeds of 327 soybean germplasm resources were determined by improved high performance liquid chromatography(HPLC).The results showed that the improved HPLC method was accurate and reliable for the determination of vitamin E in soybean.The variation coefficients of vitamin E and tocopherol contents in soybean germplasm resources were greater than 20%. The contents of α-tocopherol and total vitamin E were positively skewed, while γ and δ-tocopherol were normally distributed. The contents of α-tocopherol, γ-tocopherol, δ-tocopherol, and total vitamin E were significantly correlated. Thirty-four soybean varieties rich in vitamin E were screened by cluster analysis. Heat map analysis showed that different contents of tocopherol components of different germplasm rich in vitamin E had difference. Two germplasm resources with high content ofα-tocopherol, DongBei Black Bean and Shandongpingyi Bean, were selected. Three germplasm resources with high content ofγ-tocopherol, SCS-9299, Liaodou 22 and Shuozhoupinglu Double-color Bean were screened. Five germplasm resources with higher contents of all three tocopherols, Fuhei 2, Heizhenzhu, Yuxiuzhicuilu, Extra-large Black Testa Mung, and Qingrenhei 1were selected.
