摘  要:为了了解不同血清型鸡腺病毒(fowladenovirus,FAdV)表面蛋白的交叉反应性,本研究对FAdV-4(HN/151025株)和FAdV-11(LN/1507株)型病毒的衣壳蛋白之一的Penton蛋白分别进行原核表达以及真核表达,并分别与FAdV-11和FAdV-4型血清进行交叉反应,评价FAdV-4与FAdV-11型毒株Penton蛋白的交叉反应性。Western-blot检测结果显示,FAdV-4型的Penton蛋白与同型血清有良好的反应性,但与FAdV-11型的血清无交叉反应性;FAdV-11型的Penton蛋白与同型血清有良好的反应性,同时与FAdV-4型的血清有微弱的交叉反应。上述结果表明,FAdV-4型血清可同时识别FAdV-4型和FAdV-11型病毒的Penton蛋白。对2株病毒Penton蛋白的氨基酸序列进行比对,结果显示,两种血清型毒株的Penton蛋白的N端差异较大,这个差异区可能与蛋白的主要抗原表位区相关,进而影响血清学交叉反应性及交叉反应的强度,这可能是造成两种血清型毒株Penton蛋白交叉反应差的原因之一。To understand the cross reaction of the capsid protein between the fowl adenovirus(FAd V) of serotype 4 and 11,the Penton protein was expressed in prokaryotic and eukaryotic expression system,respectively. Western-blot was performed to evaluate the cross reaction of the Penton protein of FAd V-4 and FAd V-11 with the two types of serum. The results showed that the Penton protein of FAd V-4reacted with the same type of serum but cannot react with the serum of serotype 11.However,the Penton protein of FAd V-11 reacted with the same type serum but also was recognized by the FAd V-4 serum. The cross reaction results showed that the serum anti FAd V-4 can recognize the penton protein of FAd V-4 and FAd V-11.According to the analysis of the amino acid sequence of the two Penton proteins,the major difference located at the N terminus of the protein,which may be associated with the major epitope region and consequently influence the cross reaction extent.That may be one of the reasons that caused the poor cross reaction between two serotype viruses.
