摘 要:为了研究鼠李糖乳杆菌对妊娠后期小鼠羊水菌群多样性的影响,试验按照体重、日龄相近、配种日期相同的原则选择30只SPF级妊娠小白鼠,随机均分为对照组和灌喂组。在相同的饲养环境下,对照组小鼠灌服0.5 mL灭菌生理盐水,灌喂组小鼠灌服0.5 mL含有0.125 g鼠李糖乳杆菌的灭菌生理盐水。分娩前,在无菌条件下采集小鼠羊水,冷冻保存,用于提取小鼠羊水中细菌的RNA,分析细菌Observed_species、PD_whole_tree、Shannon、Simpson、Chao1、Ace及Goods_coverage指数,门和属水平细菌的丰度,并进行LEfSe分析。结果表明:与对照组相比,灌喂组小鼠羊水Shannon指数显著提高了20.80%(P<0.05),Simpson指数显著提高了4.26%(P<0.05);灌喂组特有的操作分类单元(operational taxonomic units, OTUs)544个;丰度较高的有厚壁菌门、变形菌门、拟杆菌门、放线菌门,且灌喂组变形菌门的丰度比对照组降低了58.64%,差异显著(P<0.05);两组间属水平的物种丰度均差异不显著(P>0.05);LEfSe分析显示,对照组的优势菌群是假单胞菌目、假单胞菌科、假单胞菌属,灌喂组是未确定的细菌门、未确定的细菌纲。说明在本试验条件下,给妊娠后期小鼠灌喂鼠李糖乳杆菌能够明显提高羊水菌群多样性,增加羊水菌种种类,改变羊水菌群丰度,显著降低变形菌门数量。In order to study the effect of Lacticaseibacillus rhamnosus on the diversity of amniotic fluid flora in mice in late pregnancy, according to the principles of body weight, similar age, and the same mating date, 30 SPF-grade pregnant mice were randomly divided into two groups, the control group and the experimental group. In the same rearing environment, the mice in the control group were fed 0.5 mL of sterilized saline, and the mice in the gavage group were fed 0.5 mL of sterilized saline containing 0.125 g of Lacticaseibacillus rhamnosus. Before delivery, mouse amniotic fluid was collected under sterile conditions, which was cryopreserved for extraction of RNA of bacteria in mouse amniotic fluid and analysis of bacterial Observed_species, PD_whole_tree, Shannon, Simpson, Chao1, Ace and Goods_coverage indexes, and abundance of bacteria at the phylum and genus levels, LEfSe was performed. The results showed that compared with control group, the Shannon index of amniotic fluid in mice in the experimental group was significantly increased by 20.80%(P<0.05), and the Simpson index was significantly increased by 4.26%(P<0.05). The OTUs(operational taxonomic units) was 544 which was unique to the experimental group;the bacteria with higher abundance were Firmicutes, Proteobacteria, Bacteroidetes, and Actinobacteria. And the abundance of the Proteobacteria in the experimental group was significantly reduced by 58.64%(P<0.05). There was no significant difference on the abundance of species at the genus level between the two groups(P>0.05). LEfSe analysis showed that the bacteria with differences in the control group were Pseudomonadaceae, Pseudomonadales, Pseudomonas;and those in the experimental group were the undetermined bacterial phylum and the undetermined bacterial class.
