摘 要:目的研究三种基源冰片对不同分子量药物经皮渗透效果的差异。方法采用改良的Franz扩散池考察三种基源冰片对小分子药物川芎嗪以及大分子药物秋水仙碱经皮渗透性的影响;利用高效液相色谱法(high performance liquid chromatography,HPLC)法测定透皮接收液中川芎嗪、秋水仙碱的含量。结果三种基源冰片对川芎嗪均具有明显的促透作用,其效果排序为合成冰片>天然冰片>艾片,但差异不显著;三种基源冰片对秋水仙碱具有更为显著的促透作用,其效果排序为天然冰片>合成冰片>艾片。结论三种基源冰片对同一药物的促透效果存在差异,说明不同化学成分与旋光性冰片对药物的促透效果存在选择性;三种基源冰片对大分子药物的促透效果更强,是可行的大分子药物经皮促透剂;合成冰片表现出不逊于天然冰片且显著优于艾片的促透效果,可替代天然冰片作为促透剂用于中药外用制剂的研发。Objective To study the transdermal permeation effects of three sourced borneol on drugs with different molecular weights.Methods Modified Franz diffusion device was used to investigate the impacts of three sourced borneol sources on the percutaneous permeability of small molecule drug ligustrazine and large molecule drug colchicine.HPLC method was used to analyze the contents of ligustrazine and colchicine in a transdermal receiving solution.Results Three sourced borneol had noticeable effects on ligustrazine penetration,and the order of effects were synthetic borneol>natural borneol>blumea camphor,but the difference was not significant;the three sourced borneol had a more substantial impact on colchicine penetration,and the order of effect was natural borneol>synthetic borneol>blumea camphor.Conclusion Three sourced borneol exhibit differences in the penetration promotion effects on the same drug,which indicates that different chemical components and optical borneol have selectivity in the penetration promotion effects of drugs;the three sourced borneol have more substantial penetration enhancement effects on macromolecular drugs and are feasible transdermal penetration enhancers for macromolecular drugs;synthetic borneol is not inferior to natural borneol but significantly superior to blumea camphor.It would be inferred that the synthetic borneol can be used as a substitute for natural borneol as a penetration enhancer for external preparations of traditional Chinese medicine.
