摘  要:采用数字基因表达谱技术(DGE),探讨成年和双侧去势大鼠附睾基因表达谱的变化。在初步筛选的2 448个差异表达基因中,与对照组相比,双侧去势组大鼠附睾中分别有1 632/816个基因表达上调/下调。qPCR验证了随机挑选的6个差异表达基因,其表达趋势均与DGE结果相符。GO分析表明差异表达基因的功能分布相似,但上调组的突触部分和辅助转运蛋白是唯一的,而电子载体和病毒粒子部分仅出现在下调组。在差异表达的基因中,有1 881个基因被定位到KEGG数据库中的250条通路。通路富集分析表明,代谢通路、氧化磷酸化通路和溶酶体通路是三条显著富集的通路。本研究将为深入研究基因在附睾中的功能提供数据支持。By using digital gene expression profiling(DGE), we investigate the differentially expressed genes in the epididymis of mature and bilateral castrated rat systematically. 2 448 candidate genes are prelimimany screered. Compared with the cantorl group, 1 632 genes are up-regulated while 816 genes are down-regulated after bilateral castration. Six differentially expressed genes are randomly selected and verifiel by qPCT, and their expression trends are consistent with the results of DGE. Using GO analysis, we find that the distributions of gene functions for up/down-regulated genes are similar, except that the synapse part and auxiliary transport protein appear to be unique in the up-regulated group, while electron carrier and virion part appear to be the only down-regulated group. Among the differentially expressed genes, 1 881 genes are mapped to 250 pathways in the KEGG database. Pathway enrichment analysis suggests that the metabolic pathway, the oxidative phosphorylation pathway and lysosome pathway are the first three significantly enriched pathways. This study will provide useful data resources for further study of gene function in epididymis.
