摘  要:目的探讨干细胞生长因子(SCF)对胰腺癌细胞生长、迁移及神经浸润的影响。方法慢病毒建立SCF过表达的胰腺癌PANC-1稳定细胞系(过表达组1)、对照细胞系(对照组1)和SCF过表达的胰腺癌AsPC-1稳定细胞系(过表达组2)、对照细胞系(对照组2),荧光定量PCR、Western blot验证各细胞系和正常胰腺导管上皮细胞HPDE6-C7中SCF mRNA、蛋白质表达。MTT法、Transwell实验分别检测转染细胞系的增殖能力、侵袭能力。体外移植瘤法构建坐骨神经浸润模型,免疫组化分析胰腺癌神经浸润情况。结果对照组1胰腺癌细胞系SCF mRNA表达(0.98±0.16)、蛋白表达(0.41±0.09)均低于过表达组1(4.32±0.78、2.18±0.37)(P<0.05);对照组2胰腺癌细胞系SCF mRNA表达(1.01±0.12)、蛋白表达(0.48±0.10)均低于过表达组2(4.85±0.51、2.58±0.31)(P<0.05);正常胰腺细胞中SCF mRNA表达(0.36±0.08)、蛋白表达(0.15±0.05)均低于对照组1、对照组2(P<0.05)。与对照组1、对照组2细胞系分别比较,过表达组1、过表达组2细胞系细胞增殖能力增加(P<0.05)。过表达组1细胞侵袭数目(85.41±12.19)高于对照组1(28.90±4.99)(P=0.001),过表达组2细胞侵袭数目(93.28±10.95)高于对照组2(34.11±6.51)(P=0.001)。对照组裸鼠坐骨神经损伤发生率[33.33%(4/12)]低于过表达组[83.33%(10/12)](χ^(2)=6.171,P=0.013)。结论SCF表达升高能促进胰腺癌PANC-1、AsPC-1细胞的生长、侵袭和神经浸润。Objective To explore the effects of stem cell growth factors(SCF)on the growth,migration and neural invasion of pancreatic cancer cells.Methods The stable pancreatic cancer PANC-1 cell line(overexpression group 1)with SCF overexpression and control cell line(control group 1),as well as stable pancreatic cancer AsPC-1 cell line(overexpression group 2)with SCF overexpression and control cell line(control group 2)were constructed by lentivirus.The expression levels of SCF mRNA and proteins in cell lines and normal pancreatic ductal epithelial cells HPDE6-C7 were verified by fluorescence quantitative PCR and Western blot.The ability and invasion abilities of transfected cell lines were detected by MTT and Transwell assay.The sciatic nerve invasion models were constructed by transplantation tumor in vitro method.The neural invasion of pancreatic cancer was analyzed by immunohistochemistry.Results The expression of SCF mRNA and protein of pancreatic cancer cell lines in control group 1 were lower than those in overexpression group 1[(0.98±0.16)and(0.41±0.09)vs.(4.32±0.78)and(2.18±0.37)](P<0.05).The expression of SCF mRNA and protein of pancreatic cancer cell lines in control group 2 were lower than those in overexpression group 2[(1.01±0.12)and(0.48±0.10)vs.(4.85±0.51)and(2.58±0.31)](P<0.05).The expression of SCF mRNA and protein in normal pancreatic cells were(0.36±0.08)and(0.15±0.05),which were lower than those in control group 1 and control group 2(P<0.05).Compared with control group 1 and control group 2,proliferation ability of cell lines were increase in overexpression group 1 and overexpression group 2(P<0.05).The number of invasion cells in overexpression group 1 was more than that in control group 1[(85.41±12.19)vs.(28.90±4.99)](P=0.001),which was more in overexpression group 2 than control group 2[(93.28±10.95)vs.(34.11±6.51)](P=0.001).The incidence of sciatic nerve injury in control group was lower than that in overexpression group[33.33%(4/12)vs.83.33%(10/12)](χ^(2)=6.171,P=0.013).Conclusion
