摘 要:目的研究石菖蒲提取物α-细辛醚缓解外周神经痛的作用机制,为研究神经损伤性疼痛、开发缓解神经性疼痛新药提供支持。方法建立大鼠坐骨神经结扎(CCI)模型。通过机械痛阈和热觉痛阈检测,观察α-细辛醚给药对CCI大鼠机械痛阈和热觉痛阈的影响。采用ELISA法检测α-细辛醚对大鼠血清IL-6、TNF-α含量的影响,采用Western blot检测α-细辛醚对大鼠脊髓磷酸化核转录因子p65、IL-6、TNF-α蛋白表达的影响。结果与对照组相比,CCI大鼠的热觉痛阈和机械痛阈均显著降低(均P<0.05),10、50 mg/kgα-细辛醚灌胃能不同程度逆转这一现象。α-细辛醚作用下CCI大鼠血清中IL-6、TNF-α质量浓度均下调(P<0.01),且呈剂量相关;CCI大鼠脊髓样本中p65、IL-6、TNF-α蛋白表达量均下调(P<0.01),且呈剂量相关。结论石菖蒲提取物α-细辛醚对外周神经痛有较好的镇痛作用,其机制可能与抑制NF-κB通路,进而抑制IL-6、TNF-α的表达有关。α-细辛醚可能会成为治疗外周神经痛的潜在候选药物。Objective To investigate the effect and mechanism ofα-asarone extracted from Acorus calamus on relieving peripheral neuralgia in rats.Methods A rat peripheral neuralgia model was established by chronic constriction injury(CCI).Through mechanical hypersensitivity and thermal hypersensitivity experiments,the effects ofα-asarone administration on mechanical and thermal pain thresholds in CCI rats were observed.The levels of serum IL 6 and TNFαin rats were detected with ELISA,and the protein expression of phosphorylated nuclear transcription factors p65,IL 6 and TNFαin rat spinal cord was detected with Western blot.Results Compared with the control group,the thermal and mechanical pain thresholds of CCI rats were significantly decreased(both P<0.05),and administration ofα-asarone 10,50 mg·kg^(-1) by gavage reversed it to varying degrees.Under the action ofα-asarone,the levels of IL 6 and TNFαin the serum of CCI rats were down-regulated(P<0.01)in a dose-dependent manner;the protein expressions of p65,IL-6 and TNF-αin the spinal cord of CCI rats were down-regulated(P<0.01),and the effect was dose-related.Conclusion Theα-asarone has a good analgesic effect on peripheral neuralgia in rats,which may be related to the inhibition of NF-κB pathway,and down-regulating the expression of IL 6 and TNFα.The study suggesting thatα-asarone may be a potential drug for the treatment of peripheral neuralgia.
