摘  要:目的分析胰岛素样生长因子-1(IGF-1)对高胆红素血症新生大鼠脑组织脑细胞凋亡的影响。方法使用腹腔注射胆红素溶液的方法成功建立40只高胆红素血症大鼠模型,将其采用随机抽样法分为4组,各10只,模型组大鼠造模后每日颈部皮下注射0.9%氯化钠溶液2 mL,小剂量组大鼠造模后每日颈部皮下注射2 mL的1μg/kg IGF-1,中剂量组大鼠造模后每日颈部皮下注射2 mL的5μg/kg IGF-1,高剂量组大鼠造模后每日颈部皮下注射2 mL的10μg/kg IGF-1。并随机纳入10只健康大鼠作为对照组,与其余各组大鼠同步每日颈部皮下注射0.9%氯化钠溶液2 mL。对各组大鼠造模后1、7、14 d的神经功能评分、脑神经细胞炎症因子水平、氧化应激水平、细胞活力、凋亡率、Bcl-2、Bax蛋白表达进行比较。结果各组大鼠造模后1 d的神经功能评分比较差异无统计学意义(P>0.05),随着时间的延长,模型组、小剂量组、中剂量组、高剂量组大鼠的神经功能评分均逐渐升高,呈现时间依赖性;造模后7、14 d时,模型组大鼠的神经功能评分均高于其他各组,且随着IGF-1水平的升高,小剂量组、中剂量组、高剂量组大鼠神经功能评分降低,呈现剂量依赖性,差异均有统计学意义(P<0.05)。随着时间的延长,模型组、小剂量组、中剂量组、高剂量组大鼠的脑神经细胞白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)、丙二醛水平、凋亡率、Bcl-2蛋白表达均逐渐升高,SOD水平、细胞活力、Bax蛋白表达均逐渐降低,呈现时间依赖性;造模后1、7、14 d时,模型组大鼠的脑神经细胞IL-6、IL-1β、丙二醛水平、凋亡率、Bcl-2蛋白表达均高于其他各组,SOD水平、细胞活力、Bax蛋白表达均低于其他各组,且随着IGF-1水平的升高,小剂量组、中剂量组、高剂量组大鼠脑神经细胞IL-6、IL-1β、丙二醛水平、凋亡率、Bcl-2蛋白表达均降低,SOD水平、细胞活力、Bax蛋白表达升高Objective To analyze the effect of insulin-like growth factor-1(IGF-1)on brain cell apoptosis in neonatal rats with hyperbilirubinemia.Methods The method of intraperitoneal injection of bilirubin solution was used to successfully establish 40 hyperbilirubinemia rat models,which were divided into 4 groups by random sampling method,each with 10 rats.The rats in the model group were necked daily after modeling.The rats in the low-dose group were injected with 2 mL of 0.9%sodium chloride solution subcutaneously.The rats in the low-dose group were injected with 2 mL of 1μg/kg IGF-1 daily in the neck after modeling,and the rats in the medium-dose group were injected with 2 mL of 5μg/kg IGF daily.The rats in the high-dose group were injected with 2 mL of 10μg/kg IGF-1 subcutaneously in the neck every day after modeling.10 healthy rats were randomly included as the control group,and 2 mL of 0.9%sodium chloride solution was injected subcutaneously into the neck simultaneously with the other groups of rats.The neurological function scores,brain nerve cell inflammatory factor levels,oxidative stress levels,cell viability,apoptosis rate,Bcl-2,Bax protein expression of 1 d,7 d,and 14 d after modeling of each group of rats were compared.Results Groups of rats after building 1 d nerve function score comparison difference was not statistically significant(P>0.05),with the extension of time,the model group,low dose group,middle dose group,high dose group rats nerve function score in a time-dependent manner;7 d and 14 d after building the model group rats nerve function score were higher than other groups,with the increase of IGF-1 level,the neurological function scores of rats in low dose group,middle dose group,high dose group decreased in a dose-dependent(P<0.05).With the prolongation of time,the levels of interleukin-6(IL-6),interleukin-1β(IL-1β),malondialdehyde(MDA),apoptosis rate and bcl-2 protein expression of brain neurons in model group,low-dose group,medium-dose group and high-dose group were gradually increased.SOD
