不同浓度氯化钾对小鼠早期胚胎体外发育的影响

摘  要:钾离子是细胞内存在的主要离子之一,在胚胎发育过程中扮演着重要角色,为了探讨钾离子对小鼠体外胚胎发育的影响,本研究将经体外受精获得的小鼠卵在分别含0 mM、2.5 mM(对照组)、12.5 mM和50 mM KCl的KSOM中进行培养。通过对其各阶段发育情况进行分析,探索钾离子浓度对小鼠早期胚胎发育的作用。结果表明,在不含KCl的KSOM中,只有极少数胚胎可以发育到2-cell时期。当KCl浓度达到12.5 mM时,发育到囊胚期的胚胎数明显减少,当KCl浓度达到50 mM时,胚胎基本都停滞在2-cell和4-cell期。此外,采用DiBAC4(3)染色方法对受精卵细胞膜电位进行了分析,结果显示高浓度的钾离子可以有效改变受精卵的细胞膜电位。本研究表明,适当浓度的钾离子是胚胎发育的前提条件之一,高浓度钾离子可影响胚胎细胞膜电位,并抑制胚胎的正常发育。Potassium ion is one of the main ions existing in cells and plays an important role in embryonic development.To investigate the effect of potassium ions on mouse embryonic development in vitro,mouse eggs obtained by in vitro fertilization were cultured in KSOM containing 0 mM,2.5 mM(control),12.5 mM and 50 mM KCl,respectively.The effect of potassium ion concentration on the early embryonic development of mice was explored by analyzing the developmental status of each stage.The results showed that in KSOM without KCl,very few embryos could develop to the 2-cell stage.When the KCl concentration reached 12.5 mM,the number of embryos that developed to the blastocyst stage was significantly reduced,and when the KCl concentration reached 50 mM,the embryos were basically arrested at the 2-and 4-cell stages.In addition,using Di-BAC4(3)staining method to analyze the membrane potential of fertilized eggs,the results show that potassium ions of high concentrations can effectively change the membrane potential of fertilized eggs.This study shows that an appropriate concentration of potassium ions is one of the prerequisites for embryonic development,and high concentrations of potassium ions can affect the embryonic cell membrane potential and inhibit the normal development of the embryo.
