摘  要:目的:针刺束缚模型抑郁大鼠,观察针刺对大鼠单胺类神经递质相关基因表达的影响。方法:领取18只雄性SD大鼠,随机分为3组:对照组、束缚组和束缚+针刺组,每组6只。采用束缚造模的方法,造模成功后进行针刺治疗。全程实验采用糖水消耗和旷野实验评价行为学。取材后进行基因检测分析。结果:束缚造模后,束缚大鼠的糖水消耗与旷野中心运动时间与对照组比较,差异具有统计学意义(P<0.01)。针刺治疗后,与对照组比较,针刺大鼠糖水消耗和旷野中心运动时间均无差异。提示针刺治疗能够逆转束缚抑郁大鼠的欣快感缺失和抑郁样行为。单胺类神经递质相关基因检测显示,针刺可以逆转抑郁大鼠以下基因:CAMP反应元件结合蛋白1(Creb1),糖原合酶激酶3α(Gsk3a),磷脂酶A2组V(Pla2g5),钙离子通道依赖电压的N型α1B亚基(Cacna1b),核受体亚家族4组A成员1(Nr4a1)。结论:针刺能够有效逆转束缚抑郁大鼠的行为并调节单胺类神经递质相关基因的表达。Objective: To observe the effect of acupuncture on the expressions of monoamine neurotransmitter-related genes in the depressed rats. Methods: The depression model was established by restraint method. 18 male SD rats were randomly divided into the control group, the model group and the acupuncture group, with 6 rats in each group. The whole experiment used sugar water consumption and field experiment to evaluate behavior. After the samples were collected, genetic testing and analysis were carried out. Results: There were statistical differences in the consumption of sugar water and the time spent in the center of field in the restrained rats compared with those in the control group(P<0.01);in which there were no statistical differences between the acupuncture group and the control group. It suggested that acupuncture treatment could reverse euphoria-absence and depression-like behaviors in restrain depressed rats. Detection of monoamine neurotransmitters-related genes showed that acupuncture could reverse the genes of CAMP response element binding protein 1(Creb1), glycogen synthase kinase 3α(Gsk3 a), phospholipase A2 group V(Pla2 g5), the voltage-dependent N-type α1 B subunit of calcium channels(Cacna1 b) and nuclear receptor subfamily 4 group A member 1(Nr4 a1) in the depressed rats. Conclusion: Acupuncture can effectively reverse the behavior of depressed rats and regulate the expression of monoamine neurotransmitter-related genes.
