一例母猪流行性腹泻的诊断

摘　　要：2020年12月份安徽省某规模猪场的妊娠母猪发生以腹泻及流产为主要症状的疾病,临床检查疑似为猪流行性腹泻。为进一步确诊本病,试验根据引起猪发生腹泻的常见4种病毒(猪流行性腹泻病毒、猪传染性胃肠炎病毒、猪轮状病毒、猪嵴病毒)的基因片段设计引物,采集妊娠流产仔猪小肠样本并提取总RNA,应用RT-PCR方法进行诊断;此外,还分别采集了妊娠30 d流产仔猪,妊娠60 d流产仔猪的胃、十二指肠、淋巴结、肾脏、心脏、脾脏、肝脏、扁桃体等内脏器官,进行组织病理学检查。结果表明:从10份流产仔猪小肠中提取的RNA均扩增得到一条特异性基因片段,与猪流行性腹泻病毒的目的基因条带大小相符;病理切片观察发现,胃黏膜层充血、水肿,十二指肠黏膜脱落和充血、出血,符合猪流行性腹泻所导致的病理变化。说明该猪场妊娠母猪感染了猪流行性腹泻病毒。In December 2020, the pregnant sows in a large-scale pig farm in Anhui Province suffered from diarrhea and abortion, which were suspected to be porcine epidemic diarrhea in clinical examination. In order to further diagnose the disease, in the experiment, primers were designed according to the gene fragments of four common viruses that caused diarrhea in pigs(Porcine epidemic diarrhea virus, Transmissible gastroenteritis virus, Porcine rotavirus, and Porcine kobuvirus);small intestine samples of pregnant and aborted piglets were collected and total RNA was extracted for diagnosis by RT-PCR method. In addition, various internal organs such as stomach, duodenum, lymph node, kidney, heart, spleen, liver, tonsil of aborted piglets on 30 and 60 days of gestation were collected for histopathological examination. The results showed that a specific gene fragment was amplified from the RNA extracted from 10 small intestine samples of aborted piglets, which was consistent with the size of the target gene band of Porcine epidemic diarrhea virus. Pathological section observation showed that the gastric mucosal layer was hyperemia and edema, and the duodenal mucosa was detached, hyperemia and hemorrhage, which were consistent with the pathological changes caused by porcine epidemic diarrhea. The results suggested that the pregnant sows in this pig farm had been infected with Porcine epidemic diarrhea virus.
