摘  要:多粘菌素E是由多粘芽孢杆菌产生的一种重要的动物抗生素,对大多数革兰氏阴性菌有较强的抗菌作用,一般通过发酵法来生产。通过控制摇瓶的装液量和玻璃珠的添加量,研究溶氧、剪切力对发酵的影响程度。在7 L罐的放大试验中,系统研究了搅拌转速、溶氧、CER、残糖等因素对效价的影响;优化流加补料工艺使得效价提高到37.5×10^(4)U/mL,实现了粘菌素E的高产。Polymyxin E is an important animal antibiotic produced by bacillus polymyxa.It has a strong antibacterial effect on most gram-negative bacteria,and is generally produced by fermentation.The effects of dissolved oxygen and shearing force on fermentation were studied by controlling the amount of liquid in the shaker flask and the amount of glass beads.In the scale-up experiment using the 7 L fermenter,the effects of stirring speed,dissolved oxygen,CER,residual sugar and other factors on titer were systematically studied.The flow and fed-feed process were further optimized to increase the titer to 37.5×10^(4)U/mL,and the high yield of polymyxin E was achieved.
