摘  要:以西红花粉末为研究对象,用超高效液相色谱质谱联用仪(ultra performance liquid chromatography-mass spectrometry,UPLC-MS)作为主要手段,确定了西红花有效成分的UPLC-MS条件。选用流动相为洗脱剂A (0.1%的甲酸水溶液,v/v),有机相为B (0.1%甲酸乙腈溶液,v/v),HSS T3柱(1.8μm×2.6 mm×100 mm)色谱柱,柱温为35℃,流速0.3 mL/min,进样量1μL, ESI离子源的正、负离子模式,正离子毛细管压为3.0 kV,负离子毛细管电压为2.0 kV。在此固定条件下,西红花溶液有效成分可以被准确地分离分析。经分析西红花中主要成分为山柰酚、藏红花酸、苦番红花素、西红花苷I和Ⅱ等10种化合物。为西红花质量控制研究奠定了基础。In this paper, the ultra performance liquid chromatography-mass spectrometry(UPLC-MS) was used to determine the conditions for the coupling of the active ingredients of saffron, mainly using saffron powder as the object of study.The mobile phase was selected as eluent A(0.1% formic acid aqueous solution, v/v), the organic phase was B(0.1% formic acid acetonitrile solution, v/v), and the HSS T3 chromatographic column(1.8 μm×2.6 mm×100 mm) was employed to UPLC analysis with the mobile phase consisting of 0.1% formic acid aqueous solution-acetonitrile. With the column temperature was 35 ℃, the flow rate was 0.3 mL/min and the injection volume was 1 μL, ESI ion source was used to ensure the data collected in positive and negative ion mode, the positive ion capillary pressure was 3.0 kV, negative ion capillary voltage was 2.0 kV. Under this fixed condition, the saffron solution can be accurately separated and analyzed. Under this fixed condition, the active ingredients of saffron solution could be accurately separated and analyzed. Ten compounds including kaempferol, saffron acid, bitter saffronin, saffron glycosides I and Ⅱ were analyzed as the main components in saffron. The method established in this study could be laid the foundation for the quality control research of saffron.
