摘  要:目的对比破伤风疫苗在BALB/c与NIH小鼠中的免疫效果,探讨将BALB/c小鼠用于破伤风疫苗效力实验的可能。方法将破伤风毒素系列稀释至适当的浓度范围,根据小鼠存活情况摸索合适的毒素浓度,重复实验,测定BALB/c和NIH小鼠的破伤风毒素半数致死量(median lethal dose,LD_(50))。分别用BALB/c和NIH小鼠的破伤风毒素2 LD_(50)同时攻击BALB/c与NIH小鼠,考察BALB/c和NIH小鼠对破伤风毒素的敏感性。将破伤风疫苗稀释50、100、200倍,分别作为高剂量组、中剂量组、低剂量组免疫BALB/c和NIH小鼠,免疫4周后用50 LD_(50)破伤风毒素攻毒,观察小鼠死亡情况。结果BALB/c小鼠的破伤风毒素2 LD_(50)为0.16μg/ml;NIH小鼠的破伤风毒素2 LD_(50)为0.23μg/ml。用0.16μg/ml的破伤风毒素分别注射BALB/c和NIH小鼠各3组,每组6只,BALB/c小鼠死亡动物数为3、3、2,NIH小鼠死亡动物数为0、0、0。用0.23μg/ml的破伤风毒素分别注射BALB/c和NIH小鼠各3组,每组6只,BALB/c小鼠死亡动物数为6、6、6,NIH小鼠死亡动物数为3、2、3。3批破伤风疫苗免疫后的BALB/c与NIH小鼠采用相同攻毒剂量,每组14只,BALB/c小鼠攻毒后,高剂量组存活数为13、14、14,中剂量组存活数为10、10、9,低剂量组存活数为4、3、4;NIH小鼠攻毒后,高剂量组存活数为10、10、10,中剂量组存活数为6、7、6,低剂量组存活数为0、0、1。结论BALB/c小鼠比NIH小鼠对破伤风毒素具有更高的敏感性,能更好地对破伤风疫苗产生免疫应答,在破伤风疫苗效价测定实验中是一种较为理想的小鼠品系。Objective To compare the immune effects of tetanus vaccine in BALB/c and NIH mice,and discuss the possibility of BALB/c mice application in tetanus vaccine efficacy experiment.Methods The tetanus toxin was diluted to proper dilution range,and the appropriate toxin concentration was explored according to the survival of mice.The experiment was repeated to determine the tetanus toxin median lethal dose(LD_(50))of BALB/c and NIH mice.BALB/c and NIH mice were challenged with 2 LD_(50) tetanus toxin of BALB/c and NIH mice,respectively,to investigate their sensitivity to tetanus toxin.Tetanus vaccine was diluted 50,100 and 200 times as high dose group,medium dose group and low dose group,respectively,to immunize BALB/c and NIH mice.Mice were challenged with 50 LD_(50) tetanus toxin 4 weeks after immunization and the death of mice was observed.Results The tetanus toxin 2 LD_(50) in BALB/c mice was 0.16μg/ml.The tetanus toxin 2 LD_(50) in NIH mice was 0.23μg/ml.Tetanus toxin at 0.16μg/ml was injected into 3 groups each of BALB/c and NIH mice,respectively,with 6 mice per group.The numbers of dead animals in BALB/c mice were 3,3,2.The numbers of dead animals in NIH mice were 0,0,0.Tetanus toxin at 0.23μg/ml was injected into 3 groups each of BALB/c and NIH mice,respectively,with 6 mice per group.The numbers of dead animals in BALB/c mice were 6,6,6.The numbers of dead animals in NIH mice were 3,2,3.BALB/c and NIH mice immunized with tetanus vaccine were treated with the same challenge dose,14 mice in each group.After BALB/c mice were challenged,the mice survival numbers of three batches of vaccines in high dose group were 13,14 and 14,those in medium dose group were 10,10 and 9,and those in low dose group were 4,3 and 4,respectively.After NIH mice were challenged,the mice survival numbers of three batches of vaccines in the high dose group were 10,10 and 10,those in the medium dose group were 6,7 and 6,and those in the low dose group were 0,0 and 1,respectively.Conclusions BALB/c mice are more sensitive to tetanus toxi
