摘 要:以草鱼鱼鳞为原料,采用四因素三水平的正交试验方法,考察超声时间、柠檬酸浓度、料液比、超声功率对鱼鳞脱钙率的影响。试验结果表明,草鱼鱼鳞脱钙最优工艺条件为超声时间80 min,柠檬酸质量分数5%,料液比1︰25 g/mL,超声功率200 W。在此条件下脱钙率可达93.8%。以羟脯氨酸溶出率作为胶原蛋白溶出影响,得此工艺下羟脯氨酸溶出率仅1.38%。因此说明超声辅助柠檬酸对草鱼鱼鳞的脱钙效果较为理想。Taking decalcification rate as index,orthogonal design of four factors three levels was taken to optimize the decalcification condition of the collagen preparation from grass carp scale by the method of citric acid with ultrasonic assistance.The main influential factors of the decalcification were ultrasonic time,concentration of the citric acid,solid-liquid ratio and ultrasonic power.The experimental resulted the most optimal condition for decalcification as follows:ultrasonic time of 80 min,citric acid concentration of 5%,solid-liquid ratio of 1︰25 g/mL and ultrasonic power of 200 W.Under this condition,the decalcification rate was up to 93.8%,but the dissolution rate of hydroxyproline was only 1.38%.It verified that the effect of decalcification by present method was a relatively ideal.
