摘  要:目的基于远志炆制前后化学成分变化情况及胃肠道毒性相关试验,探讨炆远志“减毒”的物质基础。方法采用HPLC法对生远志、制远志、炆远志中化学成分进行定量分析;采用小鼠小肠上皮隐窝IEC-6细胞系作为评价模型,以IC_(50)和丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽(GSH)为评价指标,对远志炮制前后胃肠道毒性进行评价。结果炆远志中,远志皂苷B的含量较制远志的下降程度更为显著,远志酮Ⅲ和细叶远志皂苷的含量均无明显变化,3,6′-二芥子酰基蔗糖酯的含量则缓慢下降。IEC-6细胞评价胃肠道毒性试验结果显示,综合抑制率和IC_(50)值,细胞的毒性大小为生远志>制远志>炆远志;MDA、GSH、SOD的变化情况显示,炆远志的GSH及SOD的含量均显著增加(P<0.01),MDA的含量显著降低(P<0.01),表明炆远志的毒性较制远志毒性低。结论远志经炆制,具有“减毒”的效果,具有毒性的远志皂苷类成分(如远志皂苷B)含量更低为其“减毒”原因之一。Objective To investigate the material basis of"detoxification"effect of Stewed Polygala tenuifolia based on the change of chemical compositions before and after processing and its related gastrointestinal toxicity tests.Methods HPLC was employed to quantitatively analyze the chemical compositions in crude,stewed or processed P.tenuifolia.IC_(50),MDA,SOD and GSH were adopted as evaluation indexes to assess the gastrointestinal toxicity of P.tenuifolia before and after processing using IEC-6 cell line of mouse small intestinal epithelial crypt.Results The content of polygala saponin B in stewed P.tenuifolia decreased more significantly than that in processed P.tenuifolia;polygala ketoneⅢand polygala saponin showed no obvious change,while 3,6′-diarosyl sucrose decreased slowly.The toxicity to IEC-6 cells was shown as follows based on inhibition rate and IC_(50) value:P.tenuifolia>processed P.tenuifolia>stewed P.tenuifolia,and the content of GSH and SOD of stewed P.tenuifolia increased significantly(P<0.01),while the content of MDA decreased significantly(P<0.01).In summary,the toxicity of stewed P.tenuifolia is lower than that of processed P.tenuifolia.Conclusion The stewing process showed"detoxification"effect on P.tenuifolia with one of the reason that lower content of toxic saponin(such as polygala saponin B).
