摘  要:目的采用响应面法优化紫丁香叶中总黄酮提取工艺。方法在单因素试验的基础上,以料液比、乙醇体积分数、提取时间为影响因素,总黄酮质量分数为评价指标,确定各因素水平,采用响应曲面法优化提取工艺。结果最佳提取工艺为:料液比1∶20(g∶mL),乙醇体积分数55%,提取时间50 min,提取2次,紫丁香叶总黄酮质量分数为(106.50±1.21)mg/g,与预测值107.69 mg/g接近。结论响应面法优化的最佳工艺稳定可靠,可为紫丁香叶总黄酮高效提取提供依据。Objective To optimize the extraction process of total flavonoids from Syringa oblate leaves by response surface methodology.Methods Based on single-factor experiments,the total flavonoid content was taken as the evaluation index,the ratio of solid to liquid,the volume fraction of ethanol and the extraction time were used as the influencing factors.After the level of each factor was determined,the extraction process was optimized by response surface method.Results The optimal extraction process was as follows:the ratio of solid to liquid(g∶mL)was 1∶20,the volume fraction of ethanol was 55%,the extraction time was 50 min,and the extraction times was twice.The total flavonoid content of S.oblate leaves was(106.50±1.21)mg/g,which was close to the predicted value of 107.69mg/g.Conclusion The optimal process optimized by response surface methodology is stable and reliable,which can provide a basis for efficient extraction of total flavonoids from S.oblate leaves.
