摘 要:目的研究白芍总苷(TGP)对胶原性关节炎大鼠血糖和葡萄糖转运体的调控作用。方法将30只大鼠采用随机数字表法分为正常组、模型组、TGP组和甲氨蝶呤(MTX)组。采用在大鼠右后足跖和尾根部皮下注射弗氏完全佐剂和鸡Ⅱ型胶原诱导胶原性关节炎。大鼠致炎后第14天(D14),TGP组大鼠按照每天50 mg/kg,MTX组按照每3天0.5 mg/kg,正常组和模型组给予等量溶媒,各组连续灌胃给药16 d。在大鼠致炎前和致炎后的不同时间点(D14-D30)观察大鼠足爪肿胀;大鼠致炎前和致炎后的D14和D30测量大鼠血糖浓度。末次给药(D30)结束后,酶联免疫吸附法检测血液中胰岛素水平和糖化血红蛋白水平,后处死大鼠,观察踝关节病理,蛋白质印迹法检测大鼠肝组织葡萄糖转运体-2(GLUT-2)和肌肉组织GLUT-4水平。结果与正常组相比,模型组大鼠足爪明显肿胀(P<0.01),模型组大鼠踝关节病理评分明显增高(P<0.01);与模型组相比,TGP组关节肿胀水平明显低于模型组,从D22起差异有统计学意义(P<0.01),而MTX组从D18起差异有统计学意义,TGP组与MTX组大鼠踝关节病理评分均低于模型组(P<0.01)。与正常组相比较,模型组大鼠空腹血糖水平明显升高(P<0.01),经TGP给药后,关节炎大鼠血糖水平降为(9.97±1.13)mmol/L,差异有统计学意义(P<0.01)。与正常组相比较,模型组大鼠空腹胰岛素水平与糖化血红蛋白所占总蛋白比例明显升高(P<0.01),经TGP给药后,关节炎大鼠胰岛素水平降为(16.7±1.87)μg/L,糖化血红蛋白比例降为(0.63±0.07)%,差异有统计学意义(P<0.01)。与正常组相比,模型组大鼠空腹肌糖原与肝糖原水平明显降低(P<0.01),经TGP给药后,肌糖原水平升高至(5.35±0.82)mg/g,肝糖原水平升高至(33.83±4.23)mg/g,均差异有统计学意义(P<0.01)。与正常组相比,模型组大鼠肝脏GLUT-2和肌肉组织GLUT-4水平显著下降(P<0.01);TGP给药后,GLUT-2和GLUT-4水平均显著上调,与模型组�Objective To study the regulatory effect of total glucosides of paeony(TGP)on blood glucose and glucose transporter in rats with collagen arthritis.Methods Thirty rats were randomly assigned into normal group,model group,TGP group and methotrexate(MTX)group.Collagen arthritis was induced by subcutaneous injection of Freund's complete adjuvant and chicken typeⅡcollagen at the root of the right hind foot and tail of rats.On the 14th day(D14)after inflammation,rats in TGP group were given 50 mg/kg/day and those in MTX group were given 0.5 mg/kg/3 days.The normal group and model group were given the same amount of solvent,and each group was given by gavage for 16 days.The paw swelling was observed in the rats before and at different time points after inflammation;the blood glucose was measured in the rats before and at D14 and D30 after the inflammation.After the last medicine administration(at D30),thelevels of insulin and glycosylated hemoglobin in blood were detected by enzyme-linked immunosorbent assay,and then the rats were sacrificed,and ankle pathology was observed and the levels of glucose transporter-2(GLUT-2)in liver and GLUT-4 in muscle were detected by Western blotting.Results Compared with the normal group,the paws in the model group were significantly swollen(P<0.01),and the pathological score of ankle in the model group was significantly higher(P<0.01).Compared with the model group,the level of joint swelling in TGP group was significantly lower than that in the model group;there was significant difference from D22(P<0.01),while in MTX group significant difference was seen from D18.The pathological scores of ankle joints in TGP group and MTX group were lower than that in the model group(P<0.01).Compared with the normal group,the fasting blood glucose level of the model group was significantly increased(P<0.01).After administration of TGP,the blood glucose level of arthritis rats decreased to(9.97±1.13)mmol/L;the difference was significant(P<0.01).Compared with the normal group,the fasting insulin le
