加味七方胃痛颗粒对慢性萎缩性胃炎大鼠MEK基因表达的影响

摘　　要：目的观察加味七方胃痛颗粒对慢性萎缩性胃炎大鼠模型MEK的影响。方法将120只大鼠随机分为正常组、模型组、胃复春组、加味七方胃痛颗粒大剂量、中剂量、小剂量组等6组,每组各20只。正常组自由饮用自来水,其余五组给予N-甲基-N’-硝基-N-亚硝基胍水溶液灌胃建立慢性萎缩性胃炎模型。造模成功后,正常组和模型组用生理氯化纳溶液灌胃,胃复春组、加味七方胃痛颗粒组则分别用胃复春片或相应浓度的加味七方胃痛颗粒灌胃,连续干预30天后取大鼠胃黏膜,分别用Real-time PCR、免疫组化方法以及Western blot检测MEK的表达情况。结果Real-time PCR检测MEK的基因表达显示,与模型组比较,加味七方胃痛颗粒组MEK基因表达均明显下调(P<0.01);免疫组化法检测MEK的蛋白表达显示,与模型组比较,加味七方胃痛颗粒组MEK表达均明显下调(P<0.01);Western Blot法检测MEK蛋白表达显示,与模型组比较,加味七方胃痛颗粒组MEK表达均明显下调(P<0.01)。结论加味七方胃痛颗粒对CAG有明显的疗效,其作用机制与其下调CAG大鼠的MEK基因及蛋白的表达有关。Objective To observe the effect of Jiawei Qifang Weitong Granules on MEK in rats with chronic atrophic gastritis.Methods 120 rats were randomly divided into normal group,model group,Weifuchun group,Jiawei Qifang Weitong granules high-dose,medium-dose and low-dose groups,with 20 rats in each group.The normal group was free to drink tap water,and the other five groups were given N-methyl-N’-nitrosoguanidine aqueous solution by gavage to establish the model of chronic atrophic gastritis.After successful modeling,normal group and model group were intragastrically gavaged with physiological sodium chloride solution,while Weifuchun group and Fuwei Qifang Weitong granules group were intragastrically gavaged with Weifu chun tablet and corresponding concentration of Fuwei Qifang Weitong granules.After continuous intervention for 30 days,gastric mucosa of rats was taken.Real-time PCR,immunohistochemistry and Western blot were used to detect the expression of MEK.Results Real-time PCR detection of MEK gene expression showed that,compared with model group,the expression of MEK gene in Jiawei Qifang Weitong granules group was significantly down-regulated(P<0.01).The protein expression of MEK was detected by immunohistochemical method.Compared with model group,the expression of MEK in Jiawei Qifang Weitong Granules group was significantly down-regulated(P<0.01).Western Blot analysis of MEK protein expression showed that compared with model group,MEK expression in Jiawei Qifang Weitong granules group was significantly down-regulated(P<0.01).Conclusion Jiawei Qifang Weitong Granule has obvious curative effect on CAG,and its mechanism is related to its down-regulation of MEK gene and protein expression in CAG rats.
