摘 要:目的观察三七总皂苷(Total saponins of Panax notoginseng,TPNS)对H_(2)O_(2)诱导SH-SY5Y细胞的保护作用,并探讨其作用机制。方法实验分为正常组,H_(2)O_(2)组(1000μmol/L H_(2)O_(2))和药物干预组(1000μmol·L^(-1) H_(2)O_(2)+0.1μg·mL^(-1)TPNS,1000μmol·L^(-1) H_(2)O_(2)+1μg·mL^(-1)TPNS,1000μmol·L^(-1) H_(2)O_(2)+10μg·mL^(-1)TPNS)。SH-SY5Y细胞孵育24 h后,除正常组和H_(2)O_(2)组外,其余各组加入相应的药物浓度处理12h,除正常组外,其余各组再加入相同浓度的H_(2)O_(2)处理12h。运用MTT检测细胞活力,DCFA-DA探针检测细胞内活性氧(Reactive Oxygen species,ROS)水平,JC-1检测线粒体膜电位(mitochondrial membrane potential,MMP),Western blot检测Sirt1、Foxo3a、Bcl-2和Bax蛋白的表达。结果TPNS干预组能显著提高细胞活力,降低细胞内ROS含量,改善MMP,升高Sirt1和Foxo3a蛋白表达和提高Bcl-2/Bax比值。结论TPNS可通过Sirt1/Foxo3a和Bcl-2/Bax信号通路对H_(2)O_(2)诱导SH-SY5Y细胞损伤发挥保护作用。Objective To observe the protective effect of TPNS on H_(2)O_(2)-induced SH-SY5 Y cells and its mechanism.Methods The experiment was divided into three groups:normal group,H_(2)O_(2) group(1000 mol/L H_(2)O_(2))and drug intervention group(1000μmol·L^(-1) H_(2)O_(2)+0.1μg·mL^(-1)TPNS,1000μmol·L^(-1) H_(2)O_(2)+1μg·mL^(-1)TPNS,1000μmol·L^(-1) H_(2)O_(2)+10μg·mL^(-1)TPNS).After incubation of SH-SY5 Y cells for 24 h,except the normal group and the H_(2)O_(2) group,the other groups were treated with the corresponding drug concentration for 12 h.In addition to the normal group,the other groups were treated with the same concentration of H_(2)O_(2) for 12 h.Cell viability was detected by MTT,Reactive Oxygen species(ROS)level was detected by DCFA-DA probe,mitochondrial membrane potential(MMP)was detected by JC-1,and the expression of Sirt1,Foxo3 a,bcl-2 and Bax proteins was detected by Western blot.Results The TPNS intervention group significantly increased cell activity,decreased intracellular ROS content,improved MMP,increased Sirt1 and Foxo3 a protein expression and increased bcl-2/Bax ratio.Conclusion TPNS can protect H_(2)O_(2)-induced SH-SY5 Y cells from damage through the Sirt1/Foxo3 a and bcl-2/Bax signaling pathways.
