摘 要:目的通过观察麦门冬汤对博来霉素所致BALB/C硬皮病小鼠病理影响,并探讨麦门冬汤在治疗SSc过程中防治作用及其机制。方法硬皮病小鼠模型建立后,随机将60只小鼠分为模型组、麦门冬高、中、低剂量组。并设正常对照组。治疗3周后采集标本,首先取注射破损处进行HE染色,然后摘眼球取血,同时取胸腺、脾脏和腹腔巨噬细胞,计算其脏器指数及巨噬细胞活力;用流式细胞仪检测外周血CD4^(+)、CD8^(+)T含量,ELISA法检测血清相关因子的含量。结果麦门冬汤能改善硬皮病小鼠的组织病理变化,显著影响硬皮病小鼠的胸腺指数和腹腔巨噬细胞活力(P<0.05),显著改变外周血CD4^(+)、CD8^(+)T含量,以及TGF-β、CTGF、PDGF、TNF-α、IL-4、IL-6、IL-10和INF-γ的含量(P<0.05)。结论麦门冬汤治疗SSc的机制可能与提高硬皮病模型小鼠外周血CD4^(+)、CD8^(+)T细胞水平,以及TGF-β、CTGF、PDGF、TNF-α、IL-4、IL-6、IL-10和INF-γ的含量有关。Objective Incidence of systemic sclerosis is closely related to lung and immune function.In this study,by observing the impact of Maimendong decoction in the scleroderma mice model of BALB/C,and investigate the effect and mechanism of the Wei Qi(immune function)and"Lung and fur".Methods Scleroderma mice model was established,60 mice were randomly divided into model group,the Maimendong high dose,middle dose and low dose group and normal control group.After 3 weeks of treatment,specimens were collected.First,the damaged part of the injection was taken for HE staining,and then the eyeball was taken to take blood.At the same time,the thymus,spleen and abdominal cavity macrophages were taken to calculate the organ index and macrophage viability;The contents of CD4^(+)and CD8^(+)T in peripheral blood were detected by flow cytometry,and the contents of serum related factors were detected by ELISA.Results Maimendong decoction can improve the histopathology of scleroderma mice,significantly affect the thymus index and peritoneal macrophage viability of scleroderma mice(P<0.05),and significantly change the peripheral blood CD4^(+),CD8^(+)T content,and the content of TGF-β,CTGF,PDGF,TNF-α,IL-4,IL-6,IL-10 and INF-γ(P<0.05);Conclusion The mechanism of Maimendong decoction in the treatment of scleroderma mice may be related tothe improvement of CD4^(+)and CD8^(+)T cells levels,as well as the levels of TGF-β,CTGF,PDGF,TNF-α,IL-4,IL-6,IL-10 and INF-γin peripheral blood of scleroderma model mice.
