摘  要:目的观察隔药饼灸对动脉粥样硬化易损斑块兔腹主动脉平滑肌表型转换的影响,探讨隔药饼灸稳定易损斑块的可能效应机制。方法将58只新西兰兔随机分为空白组、模型组、直接灸组、隔药饼灸组、西药组。除空白组外各组高脂饲料喂养、腹主动脉球囊损伤术、基因转染及药物触发方法建立腹主动脉粥样硬化易损斑块模型。各组采取相应干预措施。采用ELISA法检测兔血清总胆固醇(TC)、甘油三酯(TG)含量;HE染色观察兔腹主动脉血管结构;免疫组化法检测兔腹主动脉细胞外信号调节激酶1/2(ERK1/2)、α-平滑肌肌动蛋白(α-SMA)的阳性表达。结果与空白组比较,模型组血清TC、TG升高(P<0.01);与模型组比较,三组血清TC、TG降低(P<0.05或P<0.01);三组间无差异。与空白组比较,模型组ERK1/2平均光密度值升高(P<0.01);与模型组比较,三组ERK1/2平均光密度值降低(P<0.05);三组间无差异。与空白组比较,模型组α-SMA平均光密度值降低(P<0.05);与模型组比较,隔药饼灸组及西药组α-SMA平均光密度值升高(P<0.05);三组间无差异。结论隔药饼灸可稳定动脉粥样硬化易损斑块模型兔易损斑块,其机制可能与抑制血管平滑肌表型转换有关。Objective To observe the effect of Herbal-cake separated moxibustion on the phenotypic transition of abdominal aortic atherosclerosis vulnerable plaque of rabbit abdominal aortic vascular smooth muscle and to explore the possible effect mechanism of Herbal-cake separated cake moxibustion on stabilizing vulnerable plaque.Methods 58 New Zealand rabbits were randomly divided into blank group,model group,direct moxibustion group,Herbal-cake separated moxibustion group,and western medicine group.High-fat diet,abdominal aortic balloon injury surgery,gene transfection and drug triggering to establish the model of abdominal aortic atherosclerosis vulnerable plaque.Each group took corresponding intervention measures.ELISA was used to determine rabbit serum total cholesterol(TC)and triacylglycerol(TG)content.HE staining was used to observe the vascular structure of rabbit abdominal aortic plaques.Immunohistochemical method was used to detect positive expression of extracellular signal-regulated kinase 1/2(ERK1/2)andα-smooth muscle actin(α-SMA)in rabbit abdominal aorta.Results Compared with the blank group,the serum TC and TG of the model group increased(P<0.01).compared with the model group,the serum TC and TG of the three groups decreased(P<0.05 or P<0.01).there was no difference among the three groups.Compared with the blank group,the average optical density of ERK1/2 in the model group increased(P<0.01).compared with the model group,the average optical density of ERK1/2 in the three groups decreased(P<0.05).there was no difference among the three groups.Compared with the blank group,the average optical density value ofα-SMA in the model group decreased(P<0.05).compared with the model group,the average optical density value ofα-SMA increased in the medicine cake moxibustion group and the western medicine group(P<0.05).There was no difference among the groups.Conclusion Herbal-cake separated moxibustion can stabilize the vulnerable plaques in rabbits with abdominal aortic vulnerable plaque model,and its mechanism may
