摘 要:为了解贵州地方鸡群中禽白血病病毒(Avian leukosis virus,ALV)与马立克氏病毒(Marek’s disease virus,MDV)混合感染及其遗传进化情况,本研究通过对贵州威宁县某养殖场疑似禽白血病与马立克氏病混合感染的病例进行临床症状观察、解剖,并设计、合成J亚型ALV(ALV-J)gp85与MDV meq基因引物分别进行PCR检测,对扩增出的ALV-J gp85与MDV meq基因进行克隆与测序及序列分析。结果显示:临床发病鸡两腿呈劈叉状,剖检可见心脏等组织器官有肿瘤结节;ALV-J gp85与MDV meq基因的PCR扩增结果均为阳性,以上结果初步表明此病鸡属于ALV-J和MDV混合感染。对扩增出的ALV-J gp85基因进行DNA测序及序列分析,结果显示ALV-J gp85基因与ALV-J的核苷酸同源性为98.0%~99.7%,与ALV-J gp85参考毒株AHaq02核苷酸同源性高达99.7%,与ALV-J原型毒株HPRS103基因核苷酸同源性达到98.0%且位于同一分支上,与J亚群的其余参考毒株处于同一分支;MDV meq基因与超超强毒株型(vv^(+))的同源性为98.7%~99.8%,与MDV meq基因两株参考株GXY2、YLO40920同源性高达99.8%,MDV meq基因与毒力型为超超强毒株型(vv^(+))的参考毒株处于同一分支且与MDV m型、v型、vv型各代表毒株不在同一个分支上,故MDV meq基因可能属于vv^(+)型,可能为同一毒株型;以上结果进一步表明此病例为ALV-J与MDV混合感染,本研究将为肉蛋兼用型鸡ALV-J与MDV混合感染的疫情防控提供重要参考依据。In order to understand the mixed infection and its genetic evolution of Avian leukosis virus(ALV)and Marek’s disease virus(MDV)in local flocks in Guizhou,this paper conducted a survey of suspected poultry in a farm in Weining County,Guizhou Province.The mixed infection case of leukemia and Marek’s disease were clinically observed and dissected,and the primers of subtype J ALV(ALV-J)gp85 and MDV meq genes were designed and synthesized for PCR detection respectively.The amplified ALV-J gp85 and MDV meq gene was cloned and sequenced.The results showed that the legs of the clinically affected chicken were split-like,and there were tumor nodules on autopsy in the heart and other tissues and organs.The PCR amplification results of ALV-J gp85 and MDV meq genes were both positive,and the above results preliminarily indicated that the diseased chicken belongs to mixed infection of ALV-J and MDV.DNA sequencing and sequence analysis of the amplified ALV-J gp85 gene showed that the nucleotide homology between ALV-J gp85 gene and ALV-J was 98.0%~99.7%,which was similar to the reference strain of ALV-J gp85.The nucleotide homology with the ALV-J gp85 reference strain AHaq02 was as high as 99.7%,and the nucleotide homology of the HPRS103 gene of the ALV-J prototype strain reacheed 98.0%and was located on the same branch,which is in the same branch as the other reference strains of the J subgroup.The homology between MDV meq gene and the super-virulent strain type(vv^(+))was 98.7%~99.8%,and the homology with the two reference strains GXY2 and YLO40920 of the MDV meq gene was as high as 99.8%.The The MDV meq gene was on the same branch as the reference strain whose virulence type was the hypervirulent strain type(vv^(+)),and was not on the same branch as the representative strains of MDV m-type,v-type and vv-type.Therefore,the MDV meq gene may belong to the vv^(+)type or the same strain type.The above results further indicated that this case is a mixed infection of ALV-J and MDV.This study will provide an important reference
