摘 要:在分离锦绣杜鹃叶肿病菌(Exobasidium japonicum)的过程中,同时分离到1株叶栖酵母菌。为明确该叶栖酵母菌是否为锦绣杜鹃叶肿病的病原菌,并探究该菌具有何种功能。采用形态特征与分子生物学相结合的方法鉴定叶栖酵母菌,同时进行致病性测定,并通过液质联用(LC-MS/MS)技术分析该菌株培养液中代谢产物种类。经鉴定,从锦绣杜鹃叶肿病叶上分离的叶栖酵母菌为浅色戈卢别夫氏菌(Golubevia pallescens),对锦绣杜鹃叶片无致病性,说明该菌不是锦绣杜鹃叶肿病的病原菌;代谢组分析发现,培养液含有5种维生素、8种植物生长调节物质和11种氨基酸。由此可知该菌培养液含有植物生长调节物质,可作为外源物质应用于促进植物生长发育以及缓解逆境胁迫的研究中,进而为该菌作为微生物菌剂的开发利用提供理论依据,以期为后续研究叶栖酵母菌在叶片表面作用提供参考。During the process of isolating Exobasidium japonicum causing leaf-swelling in Rhododendron pulchrum,a phyllosphere yeast strain was isolated at the same time.In order to clarify whether the yeast was the pathogen of leaf-swelling in R.pulchrum,the combination of morphological characteristics and phylogenetic analysis of the ITS and LSU gene sequences was used to identify the phyllosphere yeast strain,and the pathogenicity was determined.The types of metabolites in the culture medium of the phyllosphere yeast strain were analyzed by liquid chromatography coupled with mass spectrometry(LC-MS/MS).The results showed that the strain isolated from leaves of R.pulchrum was Golubevia pallescens,which was not the pathogen causing leaf-swelling in R.pulchrum.The liquid culture of the strain contained five vitamins,eight plant growth substances and eleven amino acids.The results provide a theoretical basis for the development and utilization of G.pallescens as microbial agents.
