摘 要:目的:研究尖叶假龙胆与扯根菜不同比例配伍对酒精性肝损伤(ALD)模型大鼠的保护作用机制。方法:80只雄性SD大鼠随机分为空白组、模型组、阳性对照组、尖叶假龙胆组、扯根菜组、尖叶假龙胆与扯根菜1∶1配伍组、尖叶假龙胆与扯根菜1∶2配伍组、尖叶假龙胆与扯根菜2∶1配伍组共8组,通过酒精灌胃法制造大鼠ALD模型。实验组大鼠给予梯度酒精灌胃,空白组给予等量生理盐水灌胃,共8周。从第5周起,按照相应分组采取灌胃法给药,给药时间与酒精灌胃间隔6 h以上。第8周后取材,肝组织HE染色观察其病理变化;酶联免疫吸附法检测大鼠血清中肿瘤坏死因子-α(TNF-α)、转化生长因子-β(TGF-β)、白介素-1(IL-1)、白介素-6(IL-6)和髓样分化因子88(MyD88)水平以及肝组织中丙二醛(MDA)、超氧化物歧化酶(SOD)、乙醇脱氢酶(ADH)、乙醛脱氢酶(ALDH)和血红素氧合酶-1(HO-1)水平。结果:实验组中尖叶假龙胆与扯根菜2∶1配伍组指标变化最为显著,与模型组相比,大鼠血清中TNF-α、TGF-β、IL-1水平降低,差异具有统计学意义(P<0.01);IL-6和MyD88水平降低,具有显著差异(P<0.05);肝组织SOD和ALDH水平升高,差异具有统计学意义(P<0.01);ADH和HO-1水平升高,具有显著差异(P<0.05);MDA水平降低,具有显著差异(P<0.05);肝组织病理形态明显改善。结论:实验组中尖叶假龙胆与扯根菜2∶1配伍效果最佳,尖叶假龙胆配伍扯根菜对ALD的保护作用机制可能是通过调节MyD88途径介导的细胞信号通路进而影响血清炎症因子水平,减轻炎症反应;并通过降低氧化产物水平,提高抗氧化物水平,减轻氧化应激损伤,进而发挥改善ALD的作用。Objective:To study the protective mechanism of different proportions of the compatibility of Gentianella acuta and Penthorum chinense Pursh.in model rats with alcoholic liver disease(ALD).Methods:80 male SD rats were randomly divided into the blank group,the model group,the positive control group,the Gentianella acuta group,the Penthorum chinense Pursh.group,the 1∶1 compatibility group,the 1∶2 compatibility group and the 2∶1 compatibility group.ALD rat model was established by alcohol gavage method.The rats in the experimental group were given gradient alcohol by gavage,and the rats in the blank group were given the same amount of normal saline for 8 weeks.From the 5th week,the rats were administered by gavage according to the corresponding groups,and the interval between the administration time and alcohol gavage was more than 6 hours.The samples were collected in the 8th week.The pathological changes of liver tissues were observed by HE staining;ELISA method was used to detect the levels of TNF-α,TGF-β,IL-1,IL-6 and MyD88 in serum and the levels of MDA,SOD,ADH,ALDH and HO-1 in liver tissues.Results:In the experimental groups,the changes of the indexes were the most significant in the 2∶1 compatibility group;compared with those in the model group,the levels of TNF-α,TGF-βand IL-1 were significantly decreased(P<0.01);the levels of IL-6 and MyD88 were decreased(P<0.05);the levels of SOD and ALDH were greatly increased(P<0.01);the levels of ADH and HO-1 were increased(P<0.05);the level of MDA was decreased(P<0.05);the pathological morphology of liver tissues was significantly improved in the 2∶1 compatibility group.Conclusion:In the experimental groups,the 2∶1 compatibility of Gentianella acuta and Penthorum chinense Pursh.had the best effect.The protective mechanism of the combination in treating ALD may be through regulating the cell signaling pathway mediated by MyD88 pathway,thereby affecting the levels of serum inflammatory factors,reducing the inflammatory response,decreasing the levels of oxi
