摘 要:目的:探讨常用中药补血疗效优劣并寻找其作用机制,阐述其对造血调控因子的影响。方法:SPF级KM小鼠,经210 Gy ^(60)Co一次全身照射制备血虚模型。120只小鼠随机分为12组。实验第1天,上午各中药组分别灌胃各自中药[剂量为100 mg/(kg·d)],模型组及空白组灌胃同等体积的蒸馏水;下午除空白组外,其他各组经210 Gy ^(60)Co一次全身照射造模。各组连续灌胃10 d。比较12组体重、血红蛋白(Hb)、红细胞数量(RBC)、血细胞比容(HCT)。比较12组血清EPO、IL-3水平,肾组织EPOR mRNA表达。结果:与空白组比较,模型组体重、HCT、Hb、RBC均较低(P=0.000)。与模型组比较,药物对照组体重、HCT、Hb、RBC均较高(P=0.000),各单味中药组(除桑葚组外)体重、HCT、Hb、RBC均较高(P<0.05)。与药物对照组比较,丹参组体重较低(P<0.05)。与空白组比较,模型组血清IL-3、EPO含量均高(P<0.05)。与模型组比较,阿胶、白芍、丹参、当归、龙眼肉、桑葚组血清IL-3含量均低(P<0.05)。与药物对照组比较,当归组血清IL-3含量低(P<0.05)。另外,与模型组比较,药物对照、阿胶、丹参、龙眼肉组血清EPO含量均低(P<0.05)。与药物对照组比较,阿胶组血清EPO含量低(P<0.05),大枣、当归、熟地、制首乌组血清EPO含量高(P<0.05)。与空白组比较,模型组肾组织EPOR mRNA表达低(P<0.01)。与模型组比较,药物对照组、阿胶组、白芍组、大枣组、丹参组、当归组、龙眼肉组、桑葚组、熟地组、制首乌组肾组织EPOR mRNA表达均高(P<0.05)。与药物对照组比较,阿胶、白芍、当归、熟地、制首乌组肾组织EPOR mRNA表达低(P<0.05)。结论:常用中药可以改善血虚证,其作用不是通过提升EPO、IL-3水平来实现,其对造血调控的影响机制需要进一步研究。Objective:To explore the advantages and disadvantages of the curative effect of traditional Chinese medicine on tonifying blood and to find its mechanism,and to expound its influence on hematopoietic regulatory factors.Method:120 KM mice of SPF grade were irradiated with 210 Gy ^(60)Co to establish a blood deficiency model.They were randomly divided into 12 groups.On the first day of the experiment,in the morning,each traditional Chinese medicine group was gavaged with their own traditional Chinese medicine[the dose was 100 mg/(kg·d)],and the model group and the blank group were gavaged with the same volume of distilled water;in the afternoon,except for the blank group,the other groups were exposed to a single whole-body irradiation of 210 Gy ^(60)Co.The groups were gavaged continuously for 10 d.Body weight,hemoglobin(Hb),red blood cell count(RBC)and hematocrit(HCT)were compared in 12 groups.The levels of serum EPO,IL-3 and EPOR mRNA expression in renal tissue were compared in 12 groups.Result:Compared with the blank group,the body weight,HCT,Hb and RBC of the model group were lower(P=0.000).Compared with the model group,the body weight,HCT,Hb and RBC of the drug control group were higher(P=0.000),and the body weight,HCT,Hb and RBC of each single traditional Chinese medicine group(except for Mulberry group)were higher(P<0.05).Compared with the drug control group,the body weight of Salvia Miltiorrhiza group was lower(P<0.05).Compared with the blank group,the levels of serum IL-3 and EPO in the model group were higher(P<0.05).Compared with the model group,the levels of serum IL-3 in Donkey Hide Gelatin,White Peony,Salvia Miltiorrhiza,Angelica Sinensis,Arillus Longan and Mulberry groups were all lower(P<0.05).Compared with the drug control group,the content of serum IL-3 in Angelica Sinensis group was lower(P<0.05).In addition,compared with the model group,the levels of serum EPO in the drug control,Donkey Hide Gelatin,Salvia Miltiorrhiza and Arillus Longan groups were all lower(P<0.05).Compared with the drug contr
