复合应激性ED大鼠及伊木萨克片干预后的阴茎组织miRNA差异表达谱分析

摘  要:目的筛选慢性复合应激诱发的勃起功能障碍(ED)及伊木萨克片药物干预大鼠阴茎组织差异表达的microRNA(miRNA),验证并探讨其生物学意义。方法取80只性功能正常SD雄性大鼠,从中随机抽取10只大鼠设为正常对照组,余70只为造模组,采用环境雌激素样饮食联合冷刺激建立慢性复合应激ED大鼠模型,模型建立后以APO勃起实验和交配实验从中筛选出ED大鼠,随机分为ED模型组、伊木萨克片药物干预组。药物干预3周后,提取阴茎组织总RNA进行miRNA Illumina高通量测序,筛选出ED及药物干预相关的候选差异表达miRNA,实时荧光定量PCR验证测序结果的可靠性,并进行生物信息学分析。结果筛选出ED相关miRNA候选标志物5个(4个上调,1个下调),药物干预相关miRNA候选靶点标志物8个(3个上调,5个下调)。验证结果显示,rno-miR-140-3p、rno-miR-125b-5p、rno-miR-222-3p、rno-miR-100-5p的表达与测序结果趋势一致。结论rno-miR-140-3p、rno-miR-222-3p、rno-miR-100-5p可作为ED发生及药物干预靶点相关的候选标志物,但仍需大样本量进一步验证。Objective This study aimed to screen and validate microRNAs(miRNA)in penis tissue of rat model with erectile dysfunction induced by chronic multiple stress after Yimusake tablet intervention,and to analyze their regulatory relations with possible target genes.Methods A total of 80 healthy mature male SD rats were used in this study,10 of them were selected randomly as the control group,the remainder 70 were in the model group.Chronic multiple stress rat model was established by spinach and coriander diet in the cold humid environment.Impotence rats was chosen by APO and mating experiment.Stress rats with impotence were divided into two groups:ED group and medicine intervention group.Medicine intervention was performed for 3 weeks s the rats?s penis tissue was cut to extract total RNA,and then subjected to Illumina Hiseq technology to screen differentially expressed miRNA in penis tissue of the rats,the candidate miRNA was selected and validated by qRT-PCR.Results Total of 5 differentially expressed miRNAs were selected which related to ED rats with impotence,(4 miRNAs were upregulation,1 miRNA was downregulation);8 miRNAs were selected which were related to target of medicine intervention(3 miRNAs were upregulation,5 miRNAs were downregulation).The trend of the results of rno-miR-100-5p,rno-miR-125b-5p,rno-miR-140-3p,rno-miR-222-3p were consistent with the result of high-throughput sequencing technology.Conclusion rno-miR-100-5p,rno-miR-140-3p,rno-miR-222-3p may be biomarkers in the development of erectile dysfunction,and also treated as target of medicine intervention.
