摘 要:研究旨在探讨不同处理方式给药后羊血清角质细胞生长因子2 (KGF-2)的浓度变化趋势,为重组人角质细胞生长因子2 (rhKGF-2)调控羊绒生长筛选合适的处理方法和剂量提供参考。试验选择24只5~7月龄、体重为(25.76±1.94) kg的辽宁绒山羊,公、母各半,随机分为4组,每组6个重复,每个重复1只羊。以皮下注射方式,试验1组注射rhKGF-2胶原蛋白溶液,试验2组注射rhKGF-2温敏感凝胶溶液,试验3组注射rhKGF-2生理盐水溶液,试验4组为对照组,不做rhKGF-2处理。共设3个试验周期,每个周期18 d;依次设置低、中、高(10、20、30μg/kg)等3个剂量,每个周期开始时一次性注射18 d用量;研究不同给药方式和剂量对注射后血清KGF-2浓度变化的影响,探讨适宜的给药方式和剂量。结果显示:以胶原蛋白溶液和温敏感凝胶溶液形式皮下注射rhKGF-2后,血清KGF-2浓度先增加后降低,呈近似抛物线形式变化;以生理盐水溶液形式给药后,血清KGF-2浓度迅速升高达到峰值后断崖式直线下降。随着处理剂量增加,3个试验组血清KGF-2浓度峰值出现的时间均向后延迟,KGF-2高浓度维持时间延长;在30μg/kg的剂量时,试验1组的上述两个时间参数为90和426 h,试验2组为138和426 h,试验3组为42和138 h。对于同一种处理方式,剂量增加可使血清KGF-2浓度绝对值增加。研究表明,通过外源供给方式给绒山羊补充KGF-2,增加供给剂量能够提高血清KGF-2绝对浓度、延迟峰值出现时间、延长高浓度维持时间;以胶原蛋白溶液和温敏感凝胶的方式补充KGF-2效果相近,但考虑到可操作性,以前者的效果更好。The purpose of the study was to investigate the changing trend of the concentration of KGF-2 in sheep serum after different treatments and to provide reference for screening of appropriate treatment methods and doses for rhKGF-2 to regulate cashmere growth. Twenty-four Liaoning Cashmere goats aged 5~7 months and weighing(25.76±1.94) kg, half in male and half female, were randomly divided into four groups with six repelicates in each group,and one goat in each repelicate. By subcutaneous injection, group 1 was injected with rh KGF-2 collagen solution, group 2 was injected with rh KGF-2 temperature sensitive gel solution, group 3 was injected with rh KGF-2 physiological saline solution, and group 4 was the control group without rh KGF-2 treatment. There were three periods lasted 18 d in the research which was given 10, 20, 30 μg/kg. The object of the research was finding the effects of different method and quantity of rh KGF-2 on KGF-2 serum level in Liaoning Cashmere goats, getting better method and quantity. The results showed that after subcutaneous injection of rh KGF-2 in the form of collagen solution and temperature-sensitive gel solution, the serum KGF-2 concentration first increased and then decreased, changing in an approximate parabolic form. After administration in the form of normal saline solution, the serum KGF-2 concentration increased rapidly, reached a peak, and then fell off a cliff. With the increase of the treatment dose, the peak time of serum KGF-2 concentration in the three experimental groups was delayed, and the maintenance time of high rh KGF-2 concentration was prolonged. At the dose of 30 μg/kg, the above two time parameters were 90 and 426 h in group 1,138 and 426 h in group 2, 42 and 138 h in group 3. For the same treatment, increasing the dose can increase the absolute value of serum KGF-2 concentration.
