摘　　要：目的 探讨转移相关蛋白3(MTA3)对在软骨细胞中的表达情况,对软骨细胞增殖的影响及其作用机制。方法 将C28/12软骨细胞随机分为MTA3过表达组(LV MTA3)和沉默MTA3组(siMTA3),并设空白对照组(BC)、阴性对照组(NC)。Western blot检测C28/12细胞MTA3、增殖细胞核抗原(PCNA)、Wnt/β-catenin、E钙黏蛋白(E-cadherin)水平。RT-PCR测定C28/12细胞MTA3 mRNA水平。CCK8测定C28/12细胞增殖情况。结果与BC组和NC组相比,LV-MTA3组细胞MTA3蛋白和mRNA水平提高(P<0.05),si-MTA3组细胞MTA3蛋白和mRNA水平降低(P<0.05),细胞增殖率、细胞PCNA、Wnt-catenin蛋白水平降低(P<0.05)。结论 MTA3在软骨细胞中表达,能够促进软骨细胞增殖、其机制可能与抑制Wnt/β-catenin信号通路有关。Objective and its effect on the proliferation of chondrocytes.MethodsMTA3(MTA3)and control group(MTA3/8)were randomly divided into two groups. The levels of MTA3,PCNA,Wnt/β-Catenin and E-cadherin were detected by Western blot. The level of MTA3 mRNA in C28/12 cells was measured by RT-PCR. The proliferation of C28/12 cells was measured by CCK8.ReCompared with BC group and NC group,MTA3 protein and mRNA levels in lv-mta3 group were increased(P<0.05),MTA3 protein and mRNA levels in si-mta3 group were decreased(P<0.05),cell proliferation rate,PCNA and Wnt catenin protein levels were decreased(P<0.05).Conclusion tion of chondrocytes,and its mechanism may be the inhibition of Wnt/β-Catenin The signal pathway is related.
