摘 要:建立高效液相色谱外标法测定鸢尾根茎中的鸢尾酮含量。采用C18色谱柱,流动相比例为乙腈:水=7:3,流速为1mL/min,柱温35℃,检测波长230nm。trans-α-鸢尾酮在0.0930-1.8609μg线性良好,cis-α-鸢尾酮在0.0888-1.7751μg线性良好,陈化香根鸢尾根茎中cis-α-鸢尾酮平均加样回收率为97.47%(n=9),两批陈化香根鸢尾根茎中cis-α-鸢尾酮含量分别为130.9μg/g,122.6μg/g(以干燥陈化香根鸢尾根茎计,下同)。同时,应用本方法对不同来源的鸢尾标本进行检测,测得采集于阿勒泰地区的膜苞鸢尾标本根茎中cis-α-鸢尾酮含量最高,可达177.5μg/g。故本方法可用于鸢尾根茎中鸢尾酮含量的检测;膜苞鸢尾或可作为天然来源的鸢尾酮的源植物。To establish a high performance liquid chromatography method for the determination of irone from the rhizomes of iris.By using chromatographic column of C_(18),the mobile phase ratio was acetonitrile:water=7:3 at a flow rate of 1mL/min,the column temperature was 35℃,and the detection wavelength was 230nm.In this condition,trans-α-irone has good linearity at 0.0930-1.8609μg and cis-α-irone has good linearity at 0.0888-1.7751μg.The average recovery rate of cis-α-irisone of the orris root was 97.47%(n=9).The contents of cis-α-irisone in the two batches of orris root were 130.9μg/g and 122.6μg/g(calculated as dried orris root,the same below).By using this method to detectironefrom different sources of rhizomes of iris,and it was determined that a specimens of Iris scariosa collected from the Altay areahave the highest amount of cis-α-irisone,it is reaching 177.5μg/g.This HPLC method can be used to detect the content ofirone in the rhizome of iris,and Iris scariosa can be using as a source of irone of natural produced.
