褪黑素和TGF-β信号通路与滋补肝肾中药治疗PD分子机制的关联性

摘　　要：中医认为帕金森病(Parkinson’s disease,PD)的病位在脑,主要与肝、肾等脏腑功能失调有关,临床以肝肾阴虚证型最为常见,不同的滋补肝肾中药治疗PD均有良好的效果,但其机制并不清楚。研究表明褪黑素(melatonin,MLT)和TGF-β信号通路与肝阴、肾阴可能存在关联。MLT具有抑制α-突触核蛋白(α-syn)聚集、抗氧化、抗炎、抗亚硝化、促进神经细胞生存、改善PD的非运动症状、预防DA神经元变性等作用。TGF-β信号通路影响多巴胺(dopamine,DA)神经元的发育和存活。TGF-β1保护中脑神经元免受α-syn的毒害,并具有抗凋亡、抑制小胶质细胞活化、减少炎症介质产生、促DA神经元轴突生长等作用。研究证实MLT调控TGF-β通路,而某些中药又对MLT和TGF-β通路起调控作用。滋补肝肾中药治疗PD的分子机制可能与MLT和TGF-β信号通路有关。The theory of traditional Chinese medicine(TCM)believes that Parkinson disease(PD)is located in brain,and mainly related to liver and kidney dysfunction,the most common TCM syndrome type of PD is liver-kidney yin deficiency,the use of nourishing liver and kidney herbs treating PD has a good effect,however,the mechanism is not clear.Melatonin(MLT)and TGF-βsignaling pathways may be related to liver yin and kidney yin.MLT has the functions of inhibitingα-syn aggregation,anti-oxidation,anti-inflammatory,anti-nitrosation,promoting nerve cell survival,improving non-motor symptoms of PD and preventing DA neuron degeneration.TGF-βsignaling pathway involves in the development and survival of dopamine(DA)neurons.TGF-β1 protects midbrain neurons fromα-synucleinc(α-syn)poisoning,and has the effects of anti-apoptosis,inhibiting microglia activation,reducing the production of inflammatory mediators and promoting DA neuron axonal growth.Studies have confirmed that MLT regulates TGF-βpathway,however some traditional Chinese medicines play a regulatory role on MLT and TGF-βpathways.The molecular mechanism of nourishing liver and kidney traditional Chinese medicine in the treatment of PD may be related to MLT and TGF-βsignaling pathway.
