摘 要:目的:探究刺梨多糖介导转化生长因子β(transforming growth factor-β,TGFβ)信号通路缓解运动性免疫抑制发生的信号转导机制。方法:SPF级SD大鼠40只,随机分为正常对照组(C组)、运动训练组(E组)、运动+小剂量刺梨多糖组(ES组)和运动+大剂量刺梨多糖刺梨组(EB组)。以递增负荷跑台训练模式建模免疫抑制大鼠模型,运动期间ES组、EB组分别以100 mg/kg和200 mg/kg的剂量灌胃刺梨多糖,C组、E组以等剂量的生理盐水灌胃,记录大鼠每周的体质量。试验结束后,取外周血和脾脏,观察脾脏组织结构病理变化、测定各组大鼠鼻腔液中分泌型免疫球蛋白(sIgA)的水平、外周血中CD4+、CD8+、INF-γ、IL-4的水平和脾脏组织中TGF-β1、TGF-β1受体2(TGF-βreceptor,TβR2)、Smad2/3的相对表达水平。结果:与C组相比,E组体质量、sIgA含量降低(P<0.01);脾脏组织结构病理变化明显;外周血CD4+含量降低(P<0.01)、CD4+/CD8+下降(P<0.05)、INF-γ/IL-4下降(P<0.01)、INF-γ、IL-4含量升高(P<0.01);组织TGF-β1蛋白表达升高(P<0.01)、Smad2/3蛋白表达降低(P<0.01)。与E组相比,ES、EB组体质量、sIgA含量增加(P<0.01);脾脏组织结构病理变化显著减少;外周血CD4+含量升高(P<0.05)、CD4+/CD8+升高(P<0.05)、INF-γ/IL-4升高(P<0.01)、INF-γ含量降低(P<0.05)、IL-4含量降低(P<0.01);组织TGF-β1蛋白表达降低(P<0.01)、Smad2/3蛋白表达水平升高(P<0.01),且组间存在量效关系。结论:刺梨多糖可介导TGF-β1/Smad2/3信号通路下调TGF-β1和上调Smad2/3蛋白的表达,促进T淋巴细胞的分化,调控T淋巴细胞亚群CD4+/CD8+和细胞因子INF-γ/IL-4的稳态,缓解运动性免疫抑制的发生。Objective:To explore the signal transduction mechanism of transforming growth factorβ(TGF-β)signaling pathway mediated by Rosa roxburghii polysaccharide in relieving exercise-induced immunosuppression.Methods:40 SPF SD rats were randomly divided into control group(C group),exercise group(E group),exercise low-dose rosa roxburghii polysaccharide group(ES group)and exercise high-dose Rosa roxburghii polysaccharide group(EB group)The immunosuppression rat model was established by treadmill exercise.During the exercise,ES group and EB group were given 100 mg/kg and 200 mg/kg Rosa roxburghii polysaccharide respectively,while C group and E group were given the same dose of normal saline,and the weekly weights was recorded.After the experiment,blood and spleen were taken to observe the changes of spleen tissue structure,and the level of secretory immunoglobulin(sIgA)in nasal fluid,CD4+,CD8+,INF-γ,IL-4 in blood TGF-β1,TGF-β1 receptor 2(TβR2)and Smad2/3 in spleen tissue of rats in each group were determined.Results:Compared with C group,the weight and sIgA content of rats in group E decreased(P<0.01).Changes of spleen were obvious.The levels of CD_(4+),CD_(4+)/CD_(8+),INF-γ/IL-4 decreased(P<0.05 or P<0.01),but the levels of INF-γand IL-4 increased(P<0.01).The expression of TGF-β1 increased(P<0.01),but Smad2/3 protein decreased(P<0.01).Compared with E group,the weight and sIgA content of rats in ES group and EB groups increased(P<0.01).Changes of spleen tissue structure were obviously reduced;The levels of CD_(4+),CD_(4+)/CD_(8+) and INF-γ/IL-4 increased(P<0.05 or P<0.01),but the level of INF-γdecreased and the level of IL-4 decreased(P<0.01).The expression of TGF-β1 protein decreased and Smad2/3 protein increased(P<0.01).There was a dose-effect relationship between the groups.Conclusion:Rosa roxburghii polysaccharide can mediate TGF-β1/Smad 2/3 signaling pathway to down-regulate TGF-β1 and up-regulate Smad 2/3 protein expression,promote T lymphocyte differentiation,regulate the homeostasis of T lymphocyte su
