摘 要:目的探讨槲皮素联合贯叶金丝桃提取物对脂多糖诱导小鼠抑郁样行为的影响及作用机制。方法将90只雄性清洁级C57BL/6小鼠采用随机数字表法分为对照组、模型组、槲皮素组、盐酸氟西汀、贯叶金丝桃提取物组和槲皮素联合贯叶金丝桃提取物组,每组15只。除对照组外,其余各实验组小鼠均单次注射1 mg/kg脂多糖,制备抑郁模型。对照组和模型组给予等量生理盐水灌胃,槲皮素组给予50 mg/kg槲皮素灌胃,贯叶金丝桃提取物组给予50 mg/kg贯叶金丝桃提取物灌胃,槲皮素联合贯叶金丝桃提取物组给予槲皮素联合贯叶金丝桃提取物各50 mg/kg灌胃,盐酸氟西汀组给予2.6 mg/kg盐酸氟西汀灌胃,每天1次,连续给予21 d。观察各组小鼠糖水摄入量、水迷宫实验结果,检测小鼠脑组织炎症细胞因子白介素-6(IL-6)、白介素-10(IL-10)水平及单胺类神经递质多巴胺(DA)、去甲肾上腺素(NA)水平。结果与对照组比较,模型组小鼠糖水摄入量减少[(41.29±9.73)mL比(75.87±8.73)mL,P<0.05],水迷宫实验潜伏期明显延长[(43.34±2.03)s比(19.24±2.01)s,P<0.05],水迷宫所占目标象限时间百分比降低[(22.01±0.04)%比(40.68±0.03)%,P<0.05],脑组织炎症细胞因子白介素-6(IL-6)水平显著升高[(143.98±9.43)pg/g比(81.29±9.22)pg/g,P<0.05]、白介素-10(IL-10)水平降低[(118.93±9.56)pg/g比(169.87±9.28)pg/g,P<0.05],脑组织单胺类神经递质DA、NA水平显著降低[(723.98±109.02)ng/g比(1189.87±101.18)ng/g,(612.83±63.79)ng/g比(987.38±68.82)ng/g,P均<0.05]。与模型组和各实验组比较,槲皮素联合贯叶金丝桃提取物组小鼠糖水摄入量显著增加[(72.39±8.91)mL比(41.29±9.73)mL、(50.08±9.01)mL、(51.29±10.04)mL、(65.83±8.79)mL,P均<0.05],水迷宫实验潜伏期明显缩短[(19.18±2.11)s比(43.34±2.03)s、(37.47±2.09)s、(37.12±2.13)s、(33.01±2.09)s,P均<0.05],水迷宫所占目标象限时间百分比显著增加[(40.49±0.02)%比(22.01±0.04)%、(24.9Objective To investigate the effect and mechanism of quercetin combined with hypericum perforatum extract on lipopolysaccharide-induced depression-like behavior in a mouse model.Methods Ninety clean-grade male C57BL/6 mice were divided into six groups,i.e.,control,model,quercetin,fluoxetine hydrochloride,hypericum perforatum extract,and quercetin combined with hypericum perforatum groups using a random number table(n=15).Except the control mice,all other mice were injected with 1 mg/kg lipopolysaccharide to establish the depression model,while the control and model mice were given the same amount of normal saline,the quercetin group was given 50 mg/kg quercetin,the hypericum perforatum extract group was given 50 mg/kg hypericum perforatum extract,the quercetin combined with hypericum perforatum extract group was given same amounts of quercetin and hypericum perforatum extract,and the fluoxetine hydrochloride group was given 2.6 mg/kg fluoxetine hydrochloride(all the drugs were given by gavage once a day,for 21 days).The sugar water intake and water maze test were used to assess the change in mouse behaviors.Levels of interleukin-6(IL-6),IL-10,and monoamine neurotransmitter dopamine(DA)and norepinephrine(NA)were measured in tissues using ELISA.Results Compared with the control group,the mouse sugar water intake in the model group was decreased(41.29±9.73 vs.75.87±8.73 mL;P<0.05)and the latency of the water maze test was significantly prolonged(43.34±2.03 vs.19.24±2.01 s;P<0.05),while the water maze occupied the target quadrant time was also decreased(22.01±0.04 vs.40.68±0.03%;P<0.05),the brain tissue IL-6 level was significantly induced(143.98±9.43 vs.81.29±9.22 pg/g;P<0.05),whereas levels of IL-10(118.93±9.56 vs.169.87±9.28 pg/g;P<0.05),DA(723.98±109.02 vs.1189.87±101.18 ng/g;P<0.05)and NA(612.83±63.79 vs.987.38±68.82 ng/g;P<0.05)were significantly decreased.Moreover,compared with the model and each experimental group,the mouse sugar water intake in the quercetin combined with hypericum perforatum e
