摘  要:目的借助网络药理学方法探索干姜黄芩黄连人参汤干预2型糖尿病(T2DM)胰岛素抵抗的作用机制。方法通过中药系统药理学数据库与分析平台(TCMSP)获取干姜黄芩黄连人参汤药物活性成分与靶点,采用OMIM(http://www.omim.org)、DrugBank(https://www.drugbank.ca)与Genecards(https://www.genecards.org)数据库筛选T2DM基因靶点,再利用String11.0(https://string-db.org)数据库构建干姜黄芩黄连人参汤成分与T2DM靶点蛋白互作(PPI)网络图,从而获取PPI网络图中核心靶点,最后根据Metascape(http://metascape.org/gp/index.html)平台对干姜黄芩黄连人参汤治疗T2DM进行生物学过程与通路富集分析。结果筛选出干姜黄芩黄连人参汤60种活性成分,潜在基因靶点220个,T2DM的基因靶点2062个,成分与T2DM共同靶点148个,核心作用靶点有TNF、IL-6、TP53、CASP3、IL-10、MAPK1、AKT1、VEGFA、MMP9、EGFR1、MYC、PTGS2、MAPK8、EGF、IL-1β等,其中IL-6、IL-10、IL-1β、TNF等调节免疫炎症基因靶点在PPI网络互作图中相互作用尤为明显。信号通路分析得出干姜黄芩黄连人参汤可通过NF-κB信号通路、JAK-STAT信号通路、Toll样受体信号通路等多种信号通路干预T2DM。结论干姜黄芩黄连人参汤干预T2DM的作用机制可能是通过免疫炎症途径,该复方具有多成分、多靶点以及调控多种信号通路改善T2DM胰岛素抵抗的作用特点。Objective To explore the mechanism of Ganjiang Huangqin Huanglian Renshen Decoction in improving insulin resistance in type 2 diabetes mellitus(T2DM)using the network pharmacology analysis.Methods Traditional Chinese Medicine Systems Pharmacology Database(TCMSP)was used to obtain the main active ingredients and targets of Ganjiang Huangqin Huanglian Renshen Decoction,while the OMIM(http://www.omim.org),DrugBank(https://www.drugbank.ca)and Genecards(https://www.The genecards.org database)were used to screen T2DM gene targets.The String11.0(https://string-db.org)database was used to construct the protein-protein interaction(PPI)network between the ingredients of Ganjiang Huangqin Huanglian Renshen Decoction and the T2DM targets to obtain the core targets.The biological process and pathway enrichment analysis of the treatment of T2DM by Ganjiang Huangqin Huanglian Renshen Decoction was performed according to the Metascape(http://metascape.org/gp/index.html)platform.Results There were 60 active ingredients in Ganjiang Huangqin Huanglian Renshen Decoction identified,while there were 220 gene targets of the compound and 2062 gene targets of T2DM discovered,in which there were 148 common targets of ingredients and T2DM.The core targets were TNF,IL-6,TP53,CASP3,IL-10,MAPK1,AKT1,VEGFA,MMP9,EGFR1,MYC,PTGS2,MAPK8,EGF,and IL-1β.Among them,IL-6,IL-10,IL-1β,and TNF could regulate inflammation or immunity analyzed by using the PPI network.Moreover,the Ganjiang Huangqin Huanglian Renshen Decoction could interfere with T2DM through various signaling pathways,such as the NF-κB,JAK-STAT,and Toll-like receptor signaling pathways.Conclusion The mechanism of Ganjiang Huangqin Huanglian Renshen Decoction improvement of T2DM could be through regulation of the inflammatory immune pathway and this Decoction possesses the characteristics of multi-components and multi-targets in modulation of multiple signaling pathways to alleviate insulin resistance in T2DM.
