摘 要:【目的】鉴定存于中国林业微生物菌种保藏管理中心的99株拟盘多毛孢菌株,并探究种间差异。【方法】开展联合多基因片段(ITS-tef1-tub2)分子系统学和形态学研究,并采用统计学手段对各物种的形态指标进行评估。【结果】分子系统学和形态学结果显示这些菌株为7个物种,即冷杉拟盘多毛孢(1株)、矮棕拟盘多毛孢(18株)、肯尼亚拟盘多毛孢(19株)、庐山拟盘多毛孢(38株)、南京拟盘多毛孢(3株)、葡萄牙拟盘多毛孢(13株)和桃金娘拟盘多毛孢(7株)。通过分生孢子长度、分生孢子宽度、长宽比、顶附属丝长度和底附属丝长度这5项指标中的1项或多项可有效区分此7种。【结论】研究揭示了中国林业微生物菌种保藏管理中心保存的7种拟盘多毛孢的形态学和分子系统学特征;建议重视顶附属丝长度这一形态指标在该属分类鉴定中的应用。【Objective】The aim of the present study was to identify 99 Pestalotiopsis isolates kept in China Forestry Culture Collection Center(CFCC)and to study the interspecific differences.【Methods】Isolates were identified by combined methods of phylogeny based on ITS-tef1-tub2 loci and morphology.Conidial indicators were further evaluated by statistical method.【Results】The 99 isolates were identified as P.abietis(1 isolate),P.chamaeropis(18 isolates),P.kenyana(19 isolates),P.lushanensis(38 isolates),P.nanjingensis(3 isolates),P.portugalica(13 isolates)and P.rhodomyrtus(7 isolates).These species can be validly distinguished by morphological data of conidial length and width,length width ratio,length of apical and basal appendages.【Conclusion】The present study revealed the species diversity of Pestalotiopsis isolates kept in CFCC,and morphological and phylogenetical characters were analyzed.The length of apical appendage is a useful character to separate species within Pestalotiopsis,and is recommended to be further used in the taxonomy study of this genus.
