摘 要:目的探讨瘦素在肥胖小鼠支气管哮喘模型中的作用。方法将6~8周龄健康、雌性SPF级57BL/6J小鼠分为4组:A组为肥胖哮喘组(10只)、B组为肥胖组(9只)、C组为哮喘组(12只)和D组为正常对照组(12只)。各组小鼠肺组织进行HE染色;qPCR方法检测小鼠肺组织中瘦素基因的表达,ELISA方法检测小鼠支气管肺泡灌洗液(broncho-alveolar lavage fluid,BALF)上清及外周血清中瘦素、超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)、白细胞介素-6(interleukin-6,IL-6)和肿瘤坏死因子(tumor necrosis factor,TNF-α)的水平。结果肥胖哮喘组、肥胖组、哮喘组3组小鼠肺组织中瘦素mRNA的表达量均高于正常对照组,组间比较差异有统计学意义(P<0.01),且肥胖哮喘组的表达量高于哮喘组及肥胖组,组间比较差异有统计学意义(P<0.05);肥胖哮喘组、肥胖组、哮喘组3组小鼠BALF上清及外周血清中瘦素、MDA、IL-6和TNF-α水平均高于正常对照组,SOD水平低于对照组,组间比较差异有统计学意义(P<0.05或P<0.01);肥胖哮喘组BALF上清及外周血清中瘦素、MDA、IL-6和TNF-α水平高于哮喘组及肥胖组,SOD水平低于哮喘组及肥胖组,组间比较差异有统计学意义(P<0.05或P<0.01)。结论瘦素在肥胖型哮喘中,可诱导氧化应激而发挥促炎作用。Objective To investigate the effects of leptin on bronchial asthma model in obese mice.Methods Female healthy SPF C57BL/6mice aged6-8weeks were divided into four groups:group A was obese-asthma group(10mice),group B was obese group(9mice),group C was asthma group(12mice)and group D was normal control group(12mice).The lung tissues of mice in each group were stained with HE.The expression of leptin gene in mouse lung was detected by qPCR,and levels of leptin,SOD,MPO,IL-1β,IL-6and TNF-αin bronchoalveolar lavage fluid(BALF)supernatant and peripheral serum of mice were detected by ELISA.Results The expression levels of leptin mRNA in lung tissue of the obese-asthma group,obese group and asthma group were higher than that of the normal control group(P<0.01).The expression level of leptin mRNA in the obese-asthma group was higher than those of the asthma group and obese group,and the differences among the groups were statistically significant(P<0.05).The levels of leptin,MDA,IL-6and TNF-αin BALF supernatant and peripheral serum of the obese-asthma group,obese group and asthmagroup were higher than those of the normal control group,while the level of SOD was lower than that of the control group,and the differences among the groups were statistically significant(P<0.05or P<0.01).The levels of leptin,MDA,IL-6and TNF-αin BALF supernatant and peripheral serum of the obese-asthma group were higher than those of the asthma group and obese group,while the level of SOD were lower than that of the asthma group and obese group,and the differences among the groups were statistically significant(P<0.05or P<0.01).Conclusion Leptin can induce oxidative stress and play a pro-inflammatory role in obese asthma.
