摘  要:为分离可以高效去除亚硝酸盐的反硝化细菌,并在养殖废水处理中进行应用,本实验通过富集培养、稀释涂布、划线分离、纯化、筛选,从活性污泥中获得两株反硝化能力较强的微生物菌株A和H,研究了温度、pH、溶解氧、碳源、硝酸盐浓度对菌株A和H反硝化作用的影响。结果显示:两株反硝化细菌经鉴定分别为Achromobacter xylosoxidans A和Pseudomonas stutzeri H。在含有溴麝香草酚蓝(bromothymol blue,BTB)的培养基中,菌株H在37℃,pH 6.0,摇床转速为50 r/min,在添加1%(w/v)的柠檬酸钠为碳源的培养基中,NO^(-)_(3)的去除率最高,为84.67%;菌株A在30℃,pH 6.0,静止条件下,以柠檬酸钠为碳源的培养基中,NO^(-)_(3)的去除率最高,为67.04%。A total of two denitrifying bacteria with the high efficiency in nitrite removal were isolated from the activated sludge via enriching,serious diluting and streaking on the plate in this study to isolate denitrifying bacteria using in aquacultural wastewater treatment.Factors affecting on the nitrite removal rate,such as temperature,pH,dissolved oxygen,carbon sources and nitrate concentration were investigated.The results showed that the two denitrifying bacteria,which named A and H,were identified as Achromobacter xylosoxidans A and Pseudomonas stutzeri H,respectively.The nitrite removal efficiency could be conducted at 84.67%by strain H under the conditions at 37℃,pH 6.0,50 r/min and 1%(w/v)of sodium citrate as sole carbon resource.The nitrite removal efficiency could be performed at 67.04%by the strain A at 30℃,pH 6.0,supplemented with 1%(w/v)of sodium citrate as sole carbon resource,without shaking.Applying the bacteria in nitrated reduction in aquacultural wastewater treatment would be useful.
