摘 要:目的探讨不同保存温度、保存时间血清对新型冠状病毒疫苗接种者血清IgM、IgG抗体检测的影响。方法收集98份血清样本,以当天检测的SARS-CoV-2 IgM、IgG抗体结果作为初始水平,将检测后的血清均分成13份,分别置于56℃30min后,4℃、-20℃和-70℃保存7、14、30 d后,-20℃保存60 d后,-70℃保存60、90 d后再次检测SARS-CoV-2 IgM、IgG抗体水平。结果与初始水平相比,在56℃30min、4℃(7、14、30d)、-20℃(7、14、30、60d)、-70℃(7、14、30、60、90d),SARS-CoV-2 IgG抗体水平均显著增高(P<0.01);在4℃(7、14 d)、-20℃(7d)SARS-CoV-2 IgM抗体水平差异均无统计学(P>0.05);56℃30min、4℃(30 d)、-20℃(14、30、60 d)、-70℃(7、14、30、60、90 d),SARS-CoV-2 IgM抗体水平均显著增高(P<0.01)。4℃保存7、14、30d SARS-CoV-2 IgM抗体阴性率逐渐减低。在相同天数(7 d),4℃、-20℃、-70℃SARS-CoV-2 IgM抗体阴性率也逐渐减低。结论用于SARS-CoV-2 IgM、IgG抗体检测的样本不宜长期冷藏或者冷冻保存,应尽量在短期内完成检测,否则会出现检测水平偏高。Objective To investigate effects of different storage temperature and duration on serum IgM and IgG antibodies of SARS-CoV-2 vaccinators.Methods 98 samples were collected for the study and SARS-CoV-2 IgM and IgG antibodies were measured.The detected serum samples were divided into 13 parts,stored at 56℃for 30 minutes,at 4℃,-20℃and-70℃for 7,14 and 30 days,at-20℃for 60 days and at-70℃for 60 and 90 days.Levels of SARS-CoV-2 IgM and IgG antibodies were detected again.Results Compared with the initial levels,the level of SARS-CoV-2 IgG antibody increased significantly at 56℃for 30 min,4℃for 7,14,and 30 days,-20℃for 7,14,30 and 60 days,-70℃for 7,14,30,60 and 90 days(P<0.01).There was no significant difference in the level of SARS-CoV-2 IgM antibody at 4℃for 7 and 14 days,-20℃for 7 days(P>0.05).The level of SARS-CoV-2 IgM antibody increased significantly at 56℃for 30 min,4℃for 30 days,-20℃for 14,30,and 60 days,-70℃for 7,14,30,60,and 90 days(P<0.01).The negative rate of SARS-CoV-2 IgM antibody decreased gradually after being stored at 4℃for 7,14 and 30 days.On the 7th days,the negative rate of SARS-CoV-2 IgM antibody decreased gradually at 4℃,-20℃and-70℃.Conclusion The samples used for the detection of SARS-COV-2 IgM and IgG antibodies should not be refrigerated or cryo-preserved for a long duration,and the detection should be completed within a short period as quick as possible,otherwise the detection level will be high.
