摘 要:【目的】研究谷胱甘肽过氧化物酶-5(glutathione peroxidase-5,GPx5)在成年牦牛隐睾及正常附睾组织的表达及分布特征,探索其对牦牛精子成熟及生殖机能的调节作用。【方法】选取4岁成年牦牛隐睾及正常附睾各8对,应用实时荧光定量PCR、免疫印迹、免疫组织化学SP法、免疫组织荧光染色法及IPP图像分析研究GPx5的表达及分布特征。【结果】(1)牦牛正常组附睾上皮为假复层柱状纤毛上皮,管腔内有大量精子,管腔外围结缔组织丰富;隐睾组附睾组织趋向纤维化,间质内胶原纤维增生,无正常精子。(2)GPx5在牦牛正常组附睾头部和体部表达均显著高于隐睾组(P<0.05)。(3)GPx5在2组附睾基细胞、晕细胞、管周肌样细胞、静纤毛及血管内皮细胞均有不同程度表达,正常组附睾上皮主细胞GPx5呈强阳性表达,而隐睾组主细胞无阳性表达;正常组附睾头部和体部顶端细胞GPx5呈阳性表达,尾部顶端细胞GPx5则呈强阳性,隐睾组附睾头、体和尾顶端细胞GPx5均呈强阳性表达。【结论】高原低氧环境下,牦牛隐睾附睾趋向纤维化;隐睾附睾头及附睾体GPx5显著降低,且主要表现为主细胞及基细胞的表达缺失,提示其与精子氧化应激损伤及抗氧化调节密切相关。[Purpose]To study the distribution characteristics of glutathione peroxidase-5(GPx5)in epididymis between the cryptorchidism and normal adult yak,and to discuss the regulation of GPx5 on sperm maturation and reproductive function of yak.[Methods]Eight pairs of cryptorchidism and normal epididymis of 4-year-old adult yaks were collected for analyses the expression and distribution of Gpx5 by using real-time fluorescence quantitative PCR,Western-blot,immunohistochemistry SP staining,immunohistofluorescence staining and IPP image statistics.[Results]1)The duc-tus epididymis of normal group was pseudostratified columnar ciliated epithelium and the tube wall was rich in connective tissue,but the ductus epididymis of cryptorchidism group tended to fibrosis with the collagen fibers in the wall and almost without normal sperm in the tube.2)The expression of Gpx5 in the caput and corpus of normal epididymis of yak was significantly higher than that in cryptorchidism group(P<0.05).3)GPx5 was expressed in epididymal basal cells,halo cells,peritubu-lar myoid cells,static cilia and vascular endothelial cells in two groups.GPx5 was strongly expressed in epididymal epithelial main cells in normal group,but not in cryptorchidism group.GPx5 was posit-ive in the apical cells of the caput and corpus,and strongly positive in the cauda of normal epididymis and the caput,corpus and cauda in cryptorchidism epididymis.[Conclusion]The cryptorchidic epi-didymis of yaks tend to fibrosis in high altitude and low oxygen environment.GPx5 is significantly decreased in the caput and corpus of cryptorchidism epididymis,and the expression of GPx5 is mainly decreased in the main cells and basal cells,suggesting that GPx5 is closely related to oxidative stress injury and antioxidant regulation of sperm.
