摘  要:【目的】对中国美利奴羊(新疆型)羊毛长度和产毛量性状进行全基因组关联分析(GWAS),挖掘影响这2个性状的遗传标记,揭示这些性状产生的遗传机制。【方法】以366只中国美利奴羊(新疆型)为对象,对其周岁羊(16月龄)和成年羊(30月龄)个体的羊毛长度和产毛量(污毛质量)性状进行描述性统计。提取试验羊DNA,采用Illumina OvineSNP50 BeadChip中密度芯片(包括54241个SNP位点),对周岁羊和成年羊的羊毛长度和产毛量进行GWAS。基于single-locus回归方法、permutation方法、Ovisaries Annotation Release 100和绵羊数量性状位点数据库(Sheep QTLdb)进行统计分析、基因注释和SNP验证。【结果】试验羊群体羊毛性状的平均值均在正常范围内,最大值和最小值均符合正态分析要求,可进行GWAS。从试验羊群体中共筛选出44798个SNP,每条染色体上的SNP数量为672~5150个,SNP间的距离为49.97~65.59 kb。挖掘出21个与羊毛性状相关的SNP,其中s65179.1、OAR25_10510703.1、OAR3_112640501.1和s56106.14个SNP分别位于KIF16B、RYR2、ANAPC1和DPP6基因内,并且这4个SNP位置均位于基因的编码区;其他SNP注释在FIBIN、HSD17B11、PIAS1基因附近。QTL分析结果表明,OAR25_10510703.1定位到2个与纤维长度相关的QTL和1个与污毛质量相关的QTL,OAR7_15117952.1定位到1个与纤维长度相关的QTL和1个与初级纤维直径变异系数相关的QTL,可见这2个位点对中国美利奴羊(新疆型)羊毛长度和产毛量具有显著性作用。【结论】在周岁和成年中国美利奴羊(新疆型)鉴定出了21个与羊毛长度与产毛量显著相关的SNP,揭示了中国美利奴羊纤维长度和产毛量性状的遗传结构。【Objective】This study explored the association and genetic markers of wool length and yield in Chinese Merino sheep(Xinjiang type)and revealed related genetic mechanism.【Method】Wool length and yield(quality of tainted wool)of 366 Chinese Merino sheep(Xinjiang type)at the ages of 16 months(yearling)and 30 months(adult)were analyzed.Wool length and yield of yearling and adult sheep were analyzed by GWAS using Illumina OvineNP50 BeadChip including 54241 SNP sites.Single-locus regression,permutation,Ovisaries Annotation Release 100 and Sheep QTLdb were used for statistical analysis,gene annotation and SNP verification.【Result】The average values of wool traits in the experimental sheep population were in the normal range and the maximum and minimum values were in accordance with the requirements of normal analysis,which were suitable for genome-wide association analysis.A total of 44798 SNPs were isolated from sheep with 672 to 5150 SNPs on each chromosome and distance between SNPs of 49.97 kb to 65.59 kb.Twenty-one SNPs related to wool traits were found with 4 SNPs of s65179.1,OAR25_10510703.1,OAR3_112640501.1 and s56106.1 in KIF16B,RYR2,ANAPC1 and DPP6 genes,respectively.Other SNP annotations were near FIBIN,HSD17B11 and PIAS1.QTL analysis showed that OAR25_10510703.1 was mapped to two QTLs related to fiber length and one QTL related to wool quality.OAR7_15117952.1 was mapped to one QTL related to fiber length and one QTL related to primary fiber diameter variation,indicating that these two SNPs had significant effects on wool length and yield.【Conclusion】Twenty-one SNPs associated with wool length and yield were identified in Chinese Merino sheep(Xinjiang type).This study provides a theoretical basis for revealing the genetic structure of fiber length and wool yield traits in Chinese Merino sheep.
