摘  要:目的观察复方积雪草对IgA1诱导的大鼠肾小球系膜细胞(glomerular mesangial cell,GMC)增殖及凋亡相关蛋白B细胞淋巴瘤-2(B cell lymphoma 2,Bcl-2)和胱天蛋白酶-3(caspase-3)蛋白表达的影响。方法运用高效液相色谱法检测复方积雪草煎剂溶液中主要药物的有效成分含量,按测得的比例配制复方积雪草单体组方溶液。选择合适时间和浓度的Ig A1刺激大鼠GMC,将GMC分为正常组、模型组及缬沙坦高、低浓度组、复方积雪草高、中、低浓度组,CCK-8法检测药物干预后细胞增殖情况,蛋白质印迹法检测Bcl-2、caspase-3蛋白的表达情况。结果CCK-8法检测结果显示,24h时,100μl/mlIg A1刺激GMC,模型组Ig A1刺激造模成功,模型组GMC增殖高于正常组,差异有统计学意义(P<0.05);复方积雪草高、中、低浓度组,缬沙坦高、低浓度组有明显的抑制GMC增殖作用,与模型组相比,差异均有统计学意义(P<0.05)。蛋白质印迹法检测结果显示,与正常组相比,模型组Bcl-2升高,caspase-3降低,差异均有统计学意义(P<0.05);与模型组相比,复方积雪草中浓度组、缬沙坦低浓度组的Bcl-2降低,复方积雪草中、低浓度组、缬沙坦低浓度组的caspase-3升高(P<0.05)。结论复方积雪草能明显抑制Ig A1诱导的大鼠GMC的增殖,可下调Ig A1诱导增殖后的大鼠GMC的Bcl-2蛋白表达,同时上调caspase-3蛋白表达。复方积雪草治疗IgA肾病的机制可能是通过调控细胞凋亡相关蛋白Bcl-2、caspase-3等抑制GMC的增殖。Objective To observe the effect of compound centella on rat glomerular mesangial cell(GMC)prolifera-tion induced by IgA1 and the expression of apoptosis related proteins,B cell lymphoma-2(Bcl-2)and caspase-3.Methods High performance liquid chromatography was applied to measure the effective components of main drugs in compound centella decoction,and according to the measured ratio,compound centella monomer solution was pre-pared.Rat glomerular mesangial cell were stimulated by IgA1 of certain concentration at appropriate time.GMC were divided into the normal group,model group,valsartan group with high and low concentration and compound centella group with high,medium and low concentration.CCK-8 was used to detect cell proliferation after drug intervention.The expression of Bcl-2 and caspase-3 proteins were detected by Western blotting.Results The results of the CCK-8 assay showed that the model group was successfully set by IgA1 intervention at 24 h,and the proliferation of glome-rular mesangial cell in model groupwas higher than the normal group,the difference was statistically significant(P<0.05);compound centella high,medium and low concentrationgroupand high and low concentrationvalsartan group had significant inhibition effect on glomerular mesangial cell proliferation,compared with model group,the difference were statistically significant(P<0.05).The results of Western blotting showed that,compared with normal group,Bcl-2 was increased and caspase-3 was decreased in model group,the difference were statistically significant(P<0.05);compared with model group,Bcl-2 were decreased in the compound centella medium concentration grou-pand the valsartan low concentration group,the caspase-3 was increased in the compound centella medium concen-tration group,low concentration group and the valsartan low concentration group,the difference were statistically sig-nificant(P<0.05).Conclusion Compound centella can significantly inhibit the IgA1-induced proliferation in rat GMC,downregulate the expression of Bcl-2 protein
