摘　　要：(目的)建立检测妇舒丸中正品阿胶及掺杂伪品(牛皮胶)的液相色谱-质谱方法。(方法)恒温酶解妇舒丸样品中的胶类特征性肽段,再利用液质联用多肽检测技术,对阿胶和牛皮源成分进行识别,ESI+电离模式,多响应监测模式。(结果)方法学考察中,精密度、稳定性、重复性试验RSD都小于5%;在0.1~1 mg/mL范围内,牛皮源掺入量与峰面积成线性;最低检出质量浓度约为0.01 mg/mL。用建立的方法检测妇舒丸样品,都检出阿胶且未发现牛皮源成分。(结论)该方法灵敏度高、适用性和特异性强、准确可靠,对于妇舒丸中阿胶鉴别及牛皮源限量检验具有参考意义。(Objective)An analytical method was established for the determination of donkey-hide gelatin and adulterated fake(cowhide gum)in Fushu pills.(Methods)The characteristic peptides of gelatin in the samples of Fushu pills were hydrolyzed by constant temperature enzyme,and then the components of donkey-hide gelatin and cow-hide were identified by liquid chromatography-mass spectrometry.The positive electrospray ionization and multiple reaction monitoring were adopted.(Results)RSD of precision,stability and repeatability test is less than 5%.In the range of 0.1~1 mg/mL,the mixing amount of cow leather source is linear with the peak area.The lowest detectable concentration is about 0.01 mg/mL.Donkey-hide gelatin was detected in all samples of Fushu pills by the established method,and no cowhide derived components were found.(Conclusion)The method has high sensitivity,strong applicability and specificity,accuracy and reliability.It has reference significance for the identification of donkey-hide gelatin in Fushu pills and the limit test of cow hide source.
